MAP3K1基因多態(tài)性與胃癌預(yù)后的關(guān)聯(lián)研究
[Abstract]:Objective: the whole genome association analysis (Genome-wide association studies,GWAS) showed that the single nucleotide polymorphism (single nucleotide polymorphism,SNP (SNP) of MAP3K1 rs889312 gene was a risk factor for hormone associated tumors such as breast cancer. It has been reported that hormone mediated signaling pathway plays an important role in the occurrence and development of gastric cancer. The purpose of this study is to explore the relationship between MAP3K1 rs889312 SNP and prognosis of gastric cancer. Methods: TaqMan probe method was used to detect the genotyping of 884 gastric cancer paraffin tissues after subtotal gastrectomy or total gastrectomy. Kaplan-Meier survival analysis and Cox proportional risk regression were used to analyze the polymorphism of MAP3K1 rs889312 gene. Relationship between sex and prognosis of gastric cancer. Results: our study showed that in patients with diffuse gastric cancer, MAP3K1 rs889312 heterozygous AC had a higher mortality rate than homozygous AA/CC with a higher (AC VS AA/CC log-rank P0. 028 risk ratio [hazard ratio,] HR] = 1.32 ~ 95% confidence interval [95%CI] = 1.03 ~ 1.69). In addition, univariate and multivariate Cox regression analysis showed that MAP3K1rs889312 SNP was an independent risk factor for poor prognosis in diffuse gastric cancer patients. Conclusion: SNP at MAP3K1 rs889312 site is closely related to the prognosis of patients with diffuse gastric cancer. MAP3K1 rs889312 gene polymorphism may be a molecular marker for predicting the prognosis of diffuse gastric cancer in Chinese population.
