EGCG對胃癌細胞SHH信號通路Ptch、Gli-1的表達影響

【摘要】:目的:探討(-)-Epigallocatechin gallate(EGCG)對胃癌細胞(AGS)增殖、凋亡影響,以及對Sonic hedgehog(SHH)信號通路相關(guān)蛋白Ptch、Gli-1的表達影響。方法:將AGS細胞株在37℃,5%CO2飽和濕度溫箱里進行培養(yǎng)、換液和傳代。實驗組EGCG濃度為25μg/ml、50μg/ml、100μg/ml,并設(shè)置陰性對照組,實驗時取對數(shù)期細胞。應(yīng)用MTT法和流式細胞儀檢測EGCG對AGS胃癌細胞增殖與凋亡,細胞爬片免疫組化檢測SHH信號通路相關(guān)蛋白Ptch、Gli-1的表達影響。結(jié)果:EGCG干預(yù)AGS細胞48h后,隨著EGCG濃度升高,細胞貼壁數(shù)量減少且形態(tài)變圓,漂浮數(shù)量增多,分布變稀疏,可見到細胞崩解后的碎片、顆粒等;而陰性對照組AGS細胞貼壁良好,輪廓清晰。隨著EGCG濃度增高,AGE胃癌細胞增殖抑制率、凋亡率明顯提高(P0.05);隨著EGCG濃度的升高,Gli-1表達顯著降低而Ptch表達顯著升高(P0.05)。結(jié)論:EGCG可以抑制SHH信號通路的相關(guān)蛋白促使AGS胃癌細胞凋亡
