【摘要】：目的初步探索重組人血管內(nèi)皮抑素(rh-Endostatin,rh-ES)作用于肺癌的血管正常化時相與CA9的關(guān)系,以及CA9在肺癌中的表達水平。方法收集對數(shù)生長期的Lewis細胞,制成1×106 mL-1的單細胞懸液,注射入40只C57/BL6小鼠(0.2mL/只),建立Lewis肺癌皮下移植瘤(LLC)模型,然后隨機分成對照組和rh-ES組,各20只。rh-ES組小鼠腹腔注射rh-ES 5 mg/(kg·d),9d,1次/d。對照組小鼠同時點腹腔注射生理鹽水(NS)0.2mL/次。于治療第2、4、6、9天,每組各處死5只小鼠。免疫組化檢測腫瘤組織和癌旁組織中CA9表達,Real-time PCR及ELISA技術(shù)分別檢測兩組不同時間點腫瘤組織中CA9表達。結(jié)果 C57/BL6小鼠成瘤率為100%。同時,對照組不同時間點CA9在腫瘤組織中表達均高于癌旁組織(P0.05)。Real-time PCR及ELISA發(fā)現(xiàn)在LLC移植瘤模型中CA9基因和蛋白表達在rh-ES組給藥后第4天和第6天(血管正常化時相)降低,與同組第2天、第9天比較,差異有統(tǒng)計學(xué)意義(P0.05),與對照組第4天和第6天比較,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論 CA9在腫瘤組織中高表達。rh-ES可于血管正�；瘯r間段內(nèi)降低CA9表達,逆轉(zhuǎn)Lewis肺癌乏氧。
[Abstract]:Objective to explore the relationship between recombinant human vascular endostatin (rh-Endostatin,rh-ES) and CA9 and the expression of CA9 in lung cancer. Methods Lewis cells of logarithmic growth stage were collected and single cell suspension of 1 脳 10 6 mL-1 was made. 40 C57/BL6 mice (0.2mL/ mice) were injected into (LLC) model of subcutaneous transplantation of Lewis lung cancer. Then they were randomly divided into two groups: control group and rh-ES group. The mice in rh-ES group were injected intraperitoneally with rh-ES 5 mg/ (kg d), for 9 d. Mice in control group were given intraperitoneal injection of normal saline (NS) 0.2mL/ at the same time. On the 2nd day, 5 mice died in each group. Immunohistochemical staining was used to detect the expression of CA9 in tumor tissues and paracancerous tissues. Real-time PCR and ELISA techniques were used to detect the expression of CA9 in tumor tissues at different time points. Results the tumorigenic rate of C57/BL6 mice was 100. meanwhile The expression of CA9 in tumor tissues was higher than that in adjacent tissues at different time points in control group (P0.05). The expression of CA9 gene and protein in LLC transplanted tumor model was found on the 4th and 6th day after rh-ES administration by Real-time PCR and ELISA. When the blood vessels normalize), Compared with the same group on the second day, the ninth day, the difference was statistically significant (P0.05), and compared with the control group on the 4th and 6th day, the difference was statistically significant (P0.05). Conclusion CA9 is highly expressed in tumor tissues and rh-ES can decrease the expression of CA9 and reverse hypoxia of Lewis lung cancer.
