【摘要】：核酸適體是運(yùn)用指數(shù)富集的配體系統(tǒng)進(jìn)化(systematic evolution of ligand and byexponential enrichment,SELEX)技術(shù)從人工體外合成的隨機(jī)寡核苷酸序列庫(kù)中,經(jīng)過(guò)多輪篩選后得到的單鏈DNA或RNA,具有識(shí)別靶標(biāo)范圍廣、親和力高和穩(wěn)定性好等優(yōu)勢(shì)。Cell-SELEX技術(shù)是指將整個(gè)細(xì)胞作為靶標(biāo)篩選特異性核酸適體的技術(shù),近年來(lái)已經(jīng)廣泛應(yīng)用于腫瘤細(xì)胞特異的核酸適體篩選。本研究綜述了Cell-SELEX技術(shù)的篩選過(guò)程以及通過(guò)cell-SELEX技術(shù)產(chǎn)生的核酸適體在腫瘤標(biāo)志物、腫瘤靶向治療和循環(huán)腫瘤細(xì)胞的識(shí)別與檢測(cè)等方面的應(yīng)用研究進(jìn)展。
[Abstract]:Aptamer is a random oligonucleotide sequence library synthesized in vitro using exponentially enriched ligand phylogenetic (systematic evolution of ligand and byexponential enrichment,SELEX. Single strand DNA or RNA, obtained by multiple rounds of screening has a wide range of target recognition. High affinity and good stability. Cell-SELEX technology refers to the whole cell as a target to screen specific aptamers. In recent years, it has been widely used in tumor cell specific aptamer screening. In this study, the screening process of Cell-SELEX and the application of aptamers produced by cell-SELEX in tumor markers, tumor targeting therapy and the identification and detection of circulating tumor cells were reviewed.
