龍抗Ⅰ號(hào)對(duì)放療Lewis肺癌小鼠的減毒及增效作用的研究

【摘要】:惡性腫瘤是當(dāng)前危害人類(lèi)健康的重要疾病之一,每年有大量患者死于惡性腫瘤。放療是治療惡性腫瘤的三大手段之一,是利用放射線(xiàn)照射癌細(xì)胞,讓其致死的療法。放療雖然能治療腫瘤,但可引起患者的局部放射性損傷。因此在放療的同時(shí)可以使用藥物進(jìn)行輔助治療,一方面可以減輕放療帶來(lái)的毒副作用,另一方面可以增強(qiáng)放療的治療效果,達(dá)到減毒增效的作用。目前已有多種中醫(yī)藥已經(jīng)用與輔助放療。龍抗Ⅰ號(hào)取自我國(guó)傳統(tǒng)的名貴中藥,臨床上應(yīng)用十分廣泛,與之有關(guān)的研究引起較多關(guān)注。龍抗Ⅰ號(hào)有活血化瘀、收斂止血、陣痛和抗炎的功效,并具有抗脂質(zhì)過(guò)氧化,清除氧自由基,提高超氧化物歧化酶和過(guò)氧化氫酶的活性的重要生理作用。本課題組前期研究發(fā)現(xiàn),龍抗Ⅰ號(hào)對(duì)于由重離子、中子、γ射線(xiàn)等輻射引起的大鼠損傷模型和由γ射線(xiàn)引起的骨髓型放射病小鼠模型以及γ射線(xiàn)引起的胃腸型放射病小鼠模型中,均具有一定的抗輻射作用,由此推斷龍抗Ⅰ號(hào)具有潛在的輔助放療的作用。在少量文獻(xiàn)中報(bào)道過(guò),龍抗Ⅰ號(hào)也具有一定的抗腫瘤作用。本研究繼續(xù)探究龍抗Ⅰ號(hào)對(duì)腫瘤生長(zhǎng)的影響以及對(duì)于放療的減毒及增效作用。本研究使用C57/BL小鼠成功的建立了Lewis肺癌小鼠模型,然后以預(yù)防給藥和治療給藥相結(jié)合的方式給予小鼠龍抗Ⅰ號(hào),通過(guò)對(duì)小鼠一般生活狀態(tài)、存活情況、體重變化、腫瘤體積和重量、腫瘤轉(zhuǎn)移和血象水平等多方面的考察,研究龍抗Ⅰ號(hào)對(duì)Lewis肺癌小鼠的影響。用60Coγ射線(xiàn)對(duì)Lewis肺癌小鼠進(jìn)行4Gy一次性全身輻射建立了放療Lewis肺癌小鼠模型,并以預(yù)防給藥和治療給藥相結(jié)合的方式對(duì)小鼠進(jìn)行藥物處理,通過(guò)觀(guān)察小鼠的一般生活狀態(tài)、體重變化、腫瘤體積和重量、腫瘤轉(zhuǎn)移情況和血象水平,研究龍抗Ⅰ號(hào)對(duì)放療Lewis肺癌小鼠的減毒及增效作用。同時(shí)進(jìn)行了龍抗Ⅰ號(hào)對(duì)LLC細(xì)胞存活的影響的體外實(shí)驗(yàn)。實(shí)驗(yàn)結(jié)果表明,龍抗Ⅰ號(hào)對(duì)于Lewis肺癌小鼠和放療Lewis肺癌小鼠的腫瘤生長(zhǎng)和腫瘤轉(zhuǎn)移沒(méi)明顯影響,但對(duì)Lewis肺癌小鼠和放療Lewis肺癌小鼠存活情況、一般狀態(tài)以及血象指標(biāo)有一定改善。體內(nèi)實(shí)驗(yàn)結(jié)果表明,在低劑量、短時(shí)間的處理?xiàng)l件下,龍抗Ⅰ號(hào)有一定的促進(jìn)Lewis肺癌細(xì)胞存活的作用,但隨著劑量以及時(shí)間的增加,龍抗Ⅰ號(hào)沒(méi)有表現(xiàn)出促進(jìn)或抑制Lewis肺癌細(xì)胞存活的作用,由于體內(nèi)實(shí)驗(yàn)的給藥相當(dāng)于高濃度長(zhǎng)時(shí)間的方式,所以體外實(shí)驗(yàn)與體內(nèi)實(shí)驗(yàn)的結(jié)果一致,均可說(shuō)明龍抗Ⅰ號(hào)沒(méi)有促進(jìn)Lewis肺癌細(xì)胞存活的作用。綜合以上結(jié)果得出,龍抗Ⅰ號(hào)可以作為對(duì)放療具有減毒作用的輔助用藥。
[Abstract]:Malignant tumor is one of the most important diseases endangering human health. A large number of patients die of malignant tumor every year. Radiotherapy is one of the three methods to treat malignant tumor. Although radiotherapy can treat tumor, it can cause local radiation damage in patients. Therefore, the adjuvant therapy can be carried out with drugs while radiotherapy. On the one hand, it can reduce the toxic side effects brought by radiotherapy, on the other hand, it can enhance the therapeutic effect of radiotherapy and achieve the effect of reducing toxicity and increasing efficiency. At present, a variety of Chinese medicine has been used and adjuvant radiotherapy. Longkang I is taken from Chinese traditional Chinese medicine. It is widely used in clinic, and the related research has attracted more attention. Longkang I has the functions of promoting blood circulation and removing blood stasis, astringent hemostasis, labor pain and anti-inflammation. It also has the important physiological effects of anti-lipid peroxidation, scavenging oxygen free radicals and improving the activities of superoxide dismutase and catalase. Earlier studies by our research group have found that long Kang I is used by heavy ions, neutrons, and neutrons. The damage model of rats induced by 緯-ray isradiation, the model of bone marrow radiation sickness induced by 緯-ray and the model of gastrointestinal radiation sickness induced by 緯-ray have some anti-radiation effects. It is concluded that Longkang I has the potential role of adjuvant radiotherapy. As reported in a small amount of literature, Longkang I also has some anti-tumor effect.
