【摘要】：目的:探討山茱萸提取物對原發(fā)性肝癌(hepatocellular carcinoma,HCC)大鼠B7-H6表達(dá)的影響。方法:選取60只SD大鼠隨機(jī)分為模型組、苦參堿組和山茱萸組,以二乙基亞硝胺誘發(fā)大鼠模型,分別給予苦參堿組和山茱萸組大鼠相應(yīng)藥物灌胃,模型組大鼠給予0.9%Na Cl溶液灌胃。計(jì)數(shù)各組大鼠肝臟表面的癌結(jié)節(jié)數(shù)及其抑瘤率;經(jīng)HE染色后觀察各組大鼠肝癌組織的病理學(xué)改變。應(yīng)用免疫組織化學(xué)法和Western blot法檢測模型組、苦參堿組和山茱萸組大鼠肝癌組織中B7-H6的表達(dá)。結(jié)果:苦參堿組和山茱萸組大鼠的肝癌結(jié)節(jié)數(shù)明顯少于模型組(P0.05);山茱萸組的抑瘤率顯著高于苦參堿組(P0.05)。免疫組織化學(xué)結(jié)果顯示,山茱萸組和苦參堿組肝癌組織中B7-H6的陽性表達(dá)顯著高于模型組(P0.05),山茱萸組B7-H6的陽性表達(dá)顯著高于苦參堿組(P0.05)。Western blot法結(jié)果顯示,山茱萸組和苦參堿組B7-H6的蛋白表達(dá)顯著高于模型組(P0.05),山茱萸組B7-H6的蛋白表達(dá)顯著高于苦參堿組(P0.05)。結(jié)論:山茱萸提取物可能通過上調(diào)腫瘤組織B7-H6的表達(dá),抑制大鼠肝癌組織的生長。
[Abstract]:Objective: to investigate the effect of Fructus Corni extract on the expression of B7-H6 in primary hepatic carcinoma (hepatocellular carcinoma,HCC) rats. Methods: sixty SD rats were randomly divided into three groups: model group, matrine group and Cornus officinalis group. The rats were induced by diethylnitrosamine. Rats in the model group were given 0.9%Na Cl solution. The number of tumor nodules on the liver surface and tumor inhibition rate were counted, and the pathological changes of liver cancer tissues were observed by HE staining. Immunohistochemical method and Western blot method were used to detect the expression of B7-H6 in model group, matrine group and Cornus officinalis group. Results: the number of hepatoma nodules in Matrine group and Cornus officinalis group was significantly lower than that in model group (P0.05), and the inhibitory rate in Fructus Corni group was significantly higher than that in matrine group (P0.05). Immunohistochemical results showed that the positive expression of B7-H6 in the liver cancer tissues of Cornus officinalis group and matrine group was significantly higher than that in the model group (P0.05). The positive expression of B7-H6 in Cornus officinalis group was significantly higher than that in matrine group (P0.05). Western blot method). The expression of B7-H6 protein in Cornus officinalis group and matrine group was significantly higher than that in model group (P0.05). The protein expression of B7-H6 in Cornus officinalis group was significantly higher than that in matrine group (P0.05). Conclusion: the extract of Fructus Corni may inhibit the growth of rat liver cancer by upregulating the expression of B7-H6 in tumor tissue.
