α-倒捻子素對MKN45胃癌細(xì)胞增殖、遷移、侵襲和血管生成的影響及其作用機制

【摘要】:目的:探討α-倒捻子素對人胃癌細(xì)胞MKN45增殖、遷移、侵襲和血管生成的影響及其作用機制。方法:MTS法研究α-倒捻子素對人胃癌細(xì)胞MKN45增殖的影響;傷口愈合實驗及Transwell實驗分別用于研究α-倒捻子素對MKN45細(xì)胞體外遷移及侵襲的影響;采用大鼠動脈環(huán)實驗檢測α-倒捻子素對MKN45胃癌細(xì)胞血管生成的影響;ELISA實驗檢測α-倒捻子素對MKN45細(xì)胞分泌的血管內(nèi)皮生長因子(vascular endothelial growth factor,VEGF)、腫瘤壞死因子-α(tumor necrosis factor-α,TNF-α)、轉(zhuǎn)化生長因子-β(transforming growth factor-β,TGF-β)及堿性成纖維細(xì)胞生長因子(basic fibroblast growth factor,b FGF)表達(dá)的影響等;Western blot檢測α-倒捻子素對基質(zhì)金屬蛋白酶-2(matrix metalloproteinase-2,MMP-2)、基質(zhì)金屬蛋白酶-9(matrix metalloproteinase-9,MMP-9)、神經(jīng)-鈣黏素(N-cadherin)及上皮-鈣黏素(E-cadherin)蛋白表達(dá)的影響。結(jié)果:α-倒捻子素能夠明顯抑制人胃癌細(xì)胞MKN45的增殖,且具有劑量依賴性;α-倒捻子素能夠明顯抑制MKN45細(xì)胞的遷移和侵襲;動脈環(huán)實驗結(jié)果顯示α-倒捻子素能夠抑制MKN45細(xì)胞誘導(dǎo)的血管生成。ELISA結(jié)果顯示,α-倒捻子素能夠抑制VEGF、TNF-α、TGF-β及b FGF的蛋白表達(dá);Western blot結(jié)果顯示α-倒捻子素能夠下調(diào)MMP-2、MMP-9及神經(jīng)-鈣黏素的表達(dá),同時上調(diào)上皮-鈣黏素的表達(dá)。結(jié)論:α-倒捻子素能夠抑制MKN45人胃癌細(xì)胞的增殖、遷移、侵襲和血管生成,其發(fā)揮作用可能與以下兩方面機制相關(guān):(1)抑制調(diào)控腫瘤細(xì)胞遷移及血管生成相關(guān)蛋白的表達(dá);(2)抑制MKN45細(xì)胞的上皮間質(zhì)轉(zhuǎn)化。
[Abstract]:Aim: to investigate the effects of 偽 -inverted twist on the proliferation, migration, invasion and angiogenesis of human gastric cancer cell line MKN45 and its mechanism. Methods: the effect of 偽 -inverted twist on the proliferation of human gastric cancer cell line MKN45 was studied by MTS method, and the effects of 偽 -inverted twist on the migration and invasion of MKN45 cells in vitro were studied by wound healing test and Transwell assay, respectively. The effect of 偽 -inverted twist on angiogenesis of MKN45 gastric cancer cells was detected by rat arterial ring test. ELISA assay was used to detect vascular endothelial growth factor (vascular endothelial growth factor,VEGF), tumor necrosis factor- 偽 (TNF- 偽), transforming growth factor- 尾 (TGF- 尾 (transforming growth factor- 尾) in MKN45 cells. TGF- 尾) and basic fibroblast growth factor (basic fibroblast growth factor,b FGF) expression; Western blot was used to detect matrix metalloproteinase-2 (matrix metalloproteinase-2,MMP-2), matrix metalloproteinase-9 (matrix metalloproteinase-9,MMP-9), matrix metalloproteinase-9 (matrix metalloproteinase-9,MMP-9). Effects of neurocalcitonin (N-cadherin) and epithelial-cadherin (E-cadherin) protein expression. Results: 偽 -inverted twist can inhibit the proliferation of human gastric cancer cell line MKN45 in a dose-dependent manner, and 偽 -inverted twist can inhibit the migration and invasion of MKN45 cells. The results of arterial ring experiment showed that 偽 -inverted twist could inhibit angiogenesis induced by MKN45 cells, and ELISA showed that 偽 -inverted twist could inhibit the protein expression of VEGF,TNF- 偽, TGF- 尾 and b FGF. Western blot results showed that 偽 -inverted twist could down-regulate the expression of MMP-2,MMP-9 and nerve-cadherin, and up-regulate the expression of epithelial-cadherin. Conclusion: 偽 -inverted twist can inhibit the proliferation, migration, invasion and angiogenesis of MKN45 human gastric cancer cells. Its role may be related to the following two mechanisms: (1) inhibiting the regulation of tumor cell migration and the expression of angiogenic related proteins; (2) inhibiting epithelial mesenchymal transformation of MKN45 cells.
