FTIR結(jié)合主成分分析法對(duì)直腸癌轉(zhuǎn)移淋巴結(jié)的研究

【摘要】:應(yīng)用傅里葉變換紅外光譜(FTIR)聯(lián)合主成分分析法(PCA),分析直腸癌轉(zhuǎn)移淋巴結(jié)的譜學(xué)特征,并對(duì)直腸癌轉(zhuǎn)移淋巴結(jié)和非轉(zhuǎn)移淋巴結(jié)進(jìn)行線性判別分析。80例直腸癌轉(zhuǎn)移淋巴結(jié)和80例未轉(zhuǎn)移淋巴結(jié)進(jìn)行FTIR光譜分析,計(jì)算峰強(qiáng)并進(jìn)行主成分分析,得出在波數(shù)4 000~1 700cm~(-1)范圍主成分1(Principal components 1,PC1)是3 260cm~(-1),PC2為1 740cm~(-1)。波數(shù)1 700~1 000cm~(-1)范圍,PC1為1 640cm~(-1),PC2為1 080cm~(-1),將良、惡性淋巴結(jié)光譜3 260,1 740,1 640,1 080cm~(-1)相對(duì)峰強(qiáng)比(I/I1 460)和波數(shù)1 080和1 300cm~(-1)進(jìn)行t檢驗(yàn),良、惡性結(jié)果差異有統(tǒng)計(jì)學(xué)意義(p0.05),表明癌轉(zhuǎn)移淋巴結(jié)中蛋白含量、蛋白的形成、氨基酸增多;脂肪含量明顯減少與癌組織中無(wú)氧酵解脂肪含量減少有關(guān)。將相對(duì)峰強(qiáng)比(I1 080/I1 460,I1 640/I1 460,I3 260/I1 460,I1 740/I1 460,n=160)進(jìn)行PCA聚類分析,結(jié)果顯示可以將良惡性淋巴結(jié)鑒別,良性淋巴結(jié)聚類在第一和四象限,惡性淋巴結(jié)聚類在二和三象限。將相對(duì)峰強(qiáng)比、1 080和1 300cm~(-1)進(jìn)行線性判別分析(LDA),將25例淋巴結(jié)作為驗(yàn)證集進(jìn)行分析,得出PCA/LDA模型的敏感度是87.5%,特異度是88.5%。結(jié)果表明傅里葉變換紅外光譜分析技術(shù)可成為術(shù)中原位、在體和快速診斷直腸癌淋巴結(jié)轉(zhuǎn)移的一種簡(jiǎn)便方法。
[Abstract]:Fourier transform infrared spectroscopy (FTIR) and principal component analysis (PCA),) were used to analyze the spectral characteristics of metastatic lymph nodes in rectal cancer. The metastatic lymph nodes and non-metastatic lymph nodes of rectal cancer were analyzed by linear discriminant analysis. 80 cases of metastatic lymph nodes of rectal cancer and 80 cases of non-metastatic lymph nodes were analyzed by FTIR spectrum, the peak intensity was calculated and principal component analysis was performed. It is obtained that the principal component of 1 (Principal components 1p1 is 3 260 cm-1 and PC2 is 1 740 cm-1 in the range of 4 000 ~ (-1) cm ~ (-1). The main component is 1 (Principal components ~ (-1) and the PC2 is 1 740 cm ~ (-1) in the range of 4 000 ~ (-1) cm ~ (-1). The wave number is 1 700 ~ (-1) cm ~ (-1), PC1 is 1 640 cm ~ (-1) and PC2 is 1 080 cm ~ (-1). The relative peak / intensity ratio (I/I1 460) and wave numbers of 1 080 and 1 300 cm ~ (-1) of malignant lymph nodes were measured by t test. The difference between benign and malignant results was significant (p0. 05). The results showed that the protein content, protein formation and amino acid increased in metastatic lymph nodes. The decrease of fat content was related to the decrease of anaerobic glycolytic fat content in cancer tissue. The relative peak-to-intensity ratio (I1 080/I1 460G / I1 640/I1 460N / I3 260/I1 460C / I1 740/I1 460NNX 160) was analyzed by PCA cluster analysis. The results showed that benign and malignant lymph nodes could be distinguished from benign lymph nodes in the first and fourth quadrant, and benign lymph nodes could be clustered in the first and fourth quadrants. Malignant lymph nodes were clustered in two and three quadrants. The relative peak / intensity ratio, 1 080 cm ~ (-1) and 1 300 cm ~ (-1) were analyzed by linear discriminant analysis (LDA),). The results showed that the sensitivity and specificity of the PCA/LDA model were 87.5 and 88.5respectively. The results show that Fourier transform infrared spectroscopy (FTIR) can be used as a simple method for the diagnosis of lymph node metastasis of rectal cancer in situ and in vivo.
