【摘要】：目的:探討微小RNA-138-5p(miR-138-5p)抑制肺癌細(xì)胞增殖、遷移和侵襲能力的相關(guān)機(jī)制。方法:以肺癌細(xì)胞A549和H460作為研究對象,分別轉(zhuǎn)染miR-NC(對照組)或miR-138-5p(實(shí)驗(yàn)組);生物信息學(xué)技術(shù)預(yù)測miR-138-5p的靶基因;RT-qPCR檢測轉(zhuǎn)染后細(xì)胞miR-138-5p、叉頭框蛋白C1(FOXC1)mRNA和波形蛋白(vimentin)mRNA的相對表達(dá)量;Western blot法檢測FOXC1、vimentin、E-cadherin、N-cadherin和β-catenin蛋白表達(dá)變化;MTS法和集落形成實(shí)驗(yàn)分別檢測細(xì)胞的增殖能力;劃痕愈合實(shí)驗(yàn)和Transwell法檢測細(xì)胞遷移和侵襲能力。結(jié)果:miR-138-5p過表達(dá)顯著降低FOXC1和vimentin的mRNA及蛋白的表達(dá)(P0.05),E-cadherin和β-catenin蛋白表達(dá)上調(diào),N-cadherin蛋白表達(dá)下調(diào),顯著抑制肺癌細(xì)胞的增殖、遷移和侵襲能力(P0.05)。結(jié)論:miR-138-5p可以通過靶向干擾FOXC1和vimentin的表達(dá)抑制肺癌細(xì)胞的增殖、遷移和侵襲,可能是肺癌基因治療的潛在靶點(diǎn)。
[Abstract]:Aim: to investigate the mechanism of microRNA-138-5p (miR-138-5p) inhibiting the proliferation, migration and invasion of lung cancer cells. Methods: lung cancer cells A549 and H460 were transfected with miR-NC (control group) or miR-138-5p (experimental group) respectively and the target genes of miR-138-5p were predicted by bioinformatics. The expression of FOXC1,vimentin,E-cadherin,N-cadherin and 尾-catenin protein was detected by; Western blot method after transfection with RT-qPCR. The relative expression of miR-138-5p, forkhead protein C1 (FOXC1) mRNA and vimentin (vimentin) mRNA was detected by; Western blot method. MTS assay and colony forming assay were used to detect cell proliferation ability, and scratch healing assay and Transwell assay were used to detect cell migration and invasion. Results: overexpression of miR-138-5p significantly decreased the expression of mRNA and protein in FOXC1 and vimentin (P0.05). The expression of E-cadherin and 尾 -catenin was up-regulated, and the expression of N-cadherin was down-regulated, which significantly inhibited the proliferation of lung cancer cells. Migration and invasion ability (P0.05). Conclusion: miR-138-5p can inhibit the proliferation, migration and invasion of lung cancer cells by interfering the expression of FOXC1 and vimentin, which may be a potential target of lung cancer gene therapy.
