MicroRNA在不同分化程度非小細(xì)胞肺癌中差異表達(dá)的研究

【摘要】:目的:通過芯片技術(shù)篩查不同分化程度非小細(xì)胞肺癌中Micro RNA(mi RNA)的差異表達(dá)。方法:本研究通過收集寧夏醫(yī)科大學(xué)總醫(yī)院普胸外科手術(shù)切除的肺良性病變遠(yuǎn)端正常肺組織、低分化非小細(xì)胞肺癌和高分化非小細(xì)胞肺癌組織各3例,通過提取總RNA后,使用mi RNA芯片和m RNA芯片聯(lián)合篩查,經(jīng)過篩選,找到表達(dá)差異的mi RNA。結(jié)果:經(jīng)過篩查,找到差異表達(dá)的mi RNA共27個(gè),其中相對于正常肺組織,低分化非小細(xì)胞肺癌與高分化非小細(xì)胞肺癌共同表達(dá)上調(diào)的mi RNA有9個(gè):hsa-mi R-3651,hsa-mi R-210-3p,hsa-mi R-449c-5p,hsa-mi R-375,hsa-mi R-494-3p,hsa-mi R-501-5p,hsa-mi R-664b-3p,hsa-mi R-19b-3p,hsa-mi R-200a-3p,這其中共同表達(dá)上調(diào)且分化程度越低表達(dá)上調(diào)越明顯的是:hsa-mi R-3651,hsa-mi R-210-3p。相對于正常肺組織,低分化非小細(xì)胞肺癌與高分化非小細(xì)胞肺癌中表達(dá)共同下調(diào)的有14個(gè):hsa-mi R-34c-3p,hsa-mi R-30a-3p,hsa-let-7f-5p,hsa-mi R-30c-2-3p,hsa-mi R-3136-5p,hsa-mi R-34b-3p,hsa-mi R-3911,hsa-mi R-486-3p,hsa-mi R-486-5p,hsa-mi R-6127,hsa-mi R-642a-3p,hsa-mi R-760,hsa-mi R-126-3p,hsa-mi R-1247-3p。這其中共同下調(diào)且分化程度越低下調(diào)越明顯的是:hsa-mi R-34c-3p,hsa-mi R-30a-3p。我們還發(fā)現(xiàn)了在高分化非小細(xì)胞肺癌組織內(nèi)下調(diào),低分化非小細(xì)胞肺癌組織內(nèi)上調(diào)的mi RNA有:hsa-mi R-28-5p,hsa-mi R-29b-1-5p,hsa-mi R-31-5p。高分化非小細(xì)胞肺癌組織內(nèi)上調(diào),低分化非小細(xì)胞肺癌組織內(nèi)下調(diào)的mi RNA有:hsa-mi R-449c-5p。結(jié)論:根據(jù)我們篩查到的這些mi RNA,其中hsa-mi R-3651,hsa-mi R-210-3p,hsa-mi R-34c-3p,hsa-mi R-30a-3p很有可能與非小細(xì)胞肺癌的分化程度有關(guān),而共同上調(diào)與共同下調(diào)的mi RNA很有可能是與非小細(xì)胞有關(guān)的促癌或者抑癌因子。而分化程度不同,呈現(xiàn)反向表達(dá)的hsa-mi R-28-5p,hsa-mi R-29b-1-5p,hsa-mi R-31-5p,hsa-mi R-449c-5p有可能是分化程度不同的非小細(xì)胞肺癌特有的差異mi RNA。

Methods: in this study, the distal normal lung tissues of benign lung lesions, three cases of poorly differentiated non-small cell lung cancer (NSCLC) and three cases of well-differentiated NSCLC (highly differentiated NSCLC) were collected from thoracic surgery of Ningxia Medical University General Hospital, and the total RNA was extracted. The mi RNA chip and m RNA chip are used to screen and find the mi RNA. with different expression. Results: after screening, 27 differentially expressed mi RNA were found, compared with normal lung tissue.
