【摘要】：目的:探討微小RNA(miRNA)-93在急性淋巴細(xì)胞白血病中的表達(dá)情況及其對(duì)急性T細(xì)胞白血病細(xì)胞株Jurkat增殖的影響和潛在作用機(jī)制。方法:Real-time PCR技術(shù)檢測(cè)急性白血病患者骨髓樣本中miRNA-93的表達(dá)水平。轉(zhuǎn)染miRNA-93 inhibitor下調(diào)Jurkat細(xì)胞中miRNA-93的表達(dá),分別采用CCK-8法、Ed U法和流式細(xì)胞術(shù)檢測(cè)細(xì)胞活力、增殖及周期,Western blot檢測(cè)周期相關(guān)調(diào)控因子cyclin D1、細(xì)胞周期蛋白依賴性激酶4(CDK4)、p-Rb及P27的蛋白表達(dá)水平。結(jié)果:miRNA-93在急性白血病患者中呈現(xiàn)高表達(dá),且在高�；颊咧斜磉_(dá)水平最高;沉默miRNA-93后,Jurkat細(xì)胞的活力下降并出現(xiàn)明顯的G1/S期阻滯,同時(shí)細(xì)胞中cyclin D1、CDK4、p-Rb的蛋白水平顯著降低,而P27的蛋白水平顯著升高。結(jié)論:miRNA-93在急性淋巴細(xì)胞白血病中表達(dá)顯著升高;沉默miRNA-93可通過調(diào)控周期相關(guān)因子的表達(dá)抑制急性T細(xì)胞白血病細(xì)胞株Jurkat的增殖。
[Abstract]:Aim: to investigate the expression of minimal RNA (miRNA) 93 in acute lymphoblastic leukemia (AML) and its effect on Jurkat proliferation and its potential mechanism. Methods the expression of miRNA-93 in bone marrow samples of patients with acute leukemia was detected by real-time PCR. The expression of miRNA-93 in Jurkat cells was down-regulated by transfection of miRNA-93 inhibitor, and the viability of the cells was detected by CCK-8 assay, Ed U assay and flow cytometry, respectively. The expression of cyclin dependent kinase 4 (CDK4) p-Rb and P27 protein was detected by Western blot. Results the high expression of 10 miRNA-93 was found in acute leukemia patients, and the highest expression level was found in high risk patients. After silencing miRNA-93, the activity of Jurkat cells decreased and the G 1 / S phase arrest appeared, and the protein level of cyclin D1 CDK4 pRb decreased significantly. The protein level of P27 increased significantly. Conclusion the expression of 10 miRNA-93 in acute lymphoblastic leukemia is significantly increased, and silencing miRNA-93 can inhibit the proliferation of Jurkat by regulating the expression of cycle-related factors.
