【摘要】：在女性的惡性腫瘤發(fā)病率中,乳腺癌排名第一,對女性的生命和健康造成了嚴(yán)重威脅。乳腺癌的治療包括手術(shù)治療、化療、放療、內(nèi)分泌治療及分子靶向治療。隨著對乳腺癌治療的不斷深入,多靶點(diǎn)治療的聯(lián)合應(yīng)用成為研究熱點(diǎn)。野生型p53基因是一種抑癌基因,其基因突變是乳腺癌形成的重要因素。導(dǎo)入正常的p53可以恢復(fù)p53的正常功能。重組p53腺病毒(adenovirus mediated p53 gene,Ad-p53)是將野生型p53基因?qū)胂俨《?通過其轉(zhuǎn)導(dǎo)將正常p53導(dǎo)入p53基因突變的乳腺癌細(xì)胞中,導(dǎo)入的p53合成p53蛋白,恢復(fù)p53正常的抑癌功能。雌激素受體(estrogen receptor,ER)對于乳腺癌的治療具有重要作用。乳腺癌內(nèi)分泌治療出發(fā)點(diǎn)是抑制ER發(fā)揮傳導(dǎo)信號或者降低ER數(shù)量。根據(jù)最新研究,p53和ER的表達(dá)相互調(diào)節(jié),p53可促進(jìn)ER的表達(dá);ER可抑制p53的功能。本文闡述p53、ER對乳腺癌作用機(jī)制及相互作用進(jìn)展進(jìn)行分析總結(jié),綜述有利于推動乳腺癌p53、ER雙靶點(diǎn)治療。
[Abstract]:Breast cancer ranks first in the incidence of malignant tumors in women, posing a serious threat to women's lives and health. Breast cancer treatment includes surgical treatment, chemotherapy, radiotherapy, endocrine therapy and molecular targeted therapy. With the development of breast cancer therapy, the combined application of multi-target therapy has become a hot topic. Wild type p53 gene is a tumor suppressor gene and its mutation is an important factor in the formation of breast cancer. The normal function of p53 can be restored by the introduction of normal p53. Recombinant p53 adenovirus (adenovirus mediated p53 gene,Ad-p53) is a wild type p53 gene transfected into breast cancer cells with normal p53 gene mutation through its transduction. P53 synthesizes p53 protein and recovers normal tumor suppressor function of p53. Estrogen receptor (estrogen receptor,ER) plays an important role in the treatment of breast cancer. The starting point of endocrine therapy for breast cancer is to inhibit ER signaling or reduce the number of ER. According to the latest research, the expression of p53 and ER can regulate each other. P53 can promote the expression of ER. ER can inhibit the function of p53. In this paper, the mechanism and interaction progress of p53ER in breast cancer were analyzed and summarized, which was helpful to promote the treatment of breast cancer with double target p53ER.
