IKKα及AR在前列腺癌中的表達(dá)及其臨床意義

【摘要】:目的:通過觀察IκB激酶α(inhibitor kappa B kinaseα,IKKα)及雄激素受體(androgen receptor,AR)在前列腺癌與前列腺增生中的表達(dá)變化,為進(jìn)一步探討其在前列腺癌發(fā)生、發(fā)展中的作用提供基礎(chǔ)。方法:1采用免疫組化SP法檢測(cè)30例良性前列腺增生組織與30例前列腺癌組織中IKKα及AR的表達(dá)。分析兩者在不同組織中的表達(dá)情況、兩者表達(dá)的相關(guān)性及其與前列腺癌分級(jí)、分期的關(guān)系。2采用SPSS 17.0軟件對(duì)結(jié)果進(jìn)行統(tǒng)計(jì)分析,采用卡方檢驗(yàn),并進(jìn)行秩相關(guān)分析。檢驗(yàn)標(biāo)準(zhǔn)以P0.05為有統(tǒng)計(jì)學(xué)意義。結(jié)果:1免疫組織化學(xué)結(jié)果:IKKα主要定位于腺上皮細(xì)胞的細(xì)胞漿,AR主要定位于腺上皮細(xì)胞的細(xì)胞核。IKKα在前列腺癌組中的陽(yáng)性表達(dá)率為73.33%,明顯高于前列腺增生組中的46.67%,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。AR在前列腺癌組中的陽(yáng)性表達(dá)率為83.33%,明顯高于其在前列腺增生組中的56.67%,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。2 IKKα及AR表達(dá)水平的相關(guān)性:在前列腺癌組中IKKα和AR的IHS評(píng)分二者的相關(guān)系數(shù)r=-0.533,有統(tǒng)計(jì)學(xué)意義(P0.05),說明前列腺癌組織中IKKα與AR的表達(dá)存在中等程度的負(fù)相關(guān)。3 IKKα的表達(dá)與前列腺癌分級(jí)、分期的關(guān)系:低分化前列腺癌組中IKKα的陽(yáng)性表達(dá)率為93.33%,明顯高于高分化組的53.33%,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。晚期前列腺癌組中IKKα的陽(yáng)性表達(dá)率為93.75%,明顯高于早期組的50.00%,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。4 AR的表達(dá)與前列腺癌分級(jí)、分期的關(guān)系:高分化前列腺癌組中AR的陽(yáng)性表達(dá)率為100%,明顯高于低分化組的66.67%,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。早期前列腺癌組中AR的陽(yáng)性表達(dá)率為92.86%,晚期前列腺癌組AR的陽(yáng)性表達(dá)率75.00%,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:1 IKKα及AR在前列腺癌組織中的表達(dá)陽(yáng)性率及強(qiáng)度明顯高于前列腺增生組織,說明IKKα、AR與前列腺癌有密切關(guān)系,檢測(cè)IKKα、AR有助于前列腺癌的早期診斷。2隨著前列腺癌惡性程度增加IKKα及AR的表達(dá)分別表現(xiàn)出上調(diào)和下降趨勢(shì),IKKα、AR可以作為判斷前列腺癌預(yù)后的指標(biāo)。3 IKKα與AR的表達(dá)均與前列腺癌的生物學(xué)行為、病理密切相關(guān),深入研究其作用機(jī)制,可能為前列腺癌的治療提供新的思路或方法。
[Abstract]:Aim: to investigate the expression of I 魏 B kinase 偽 (inhibitor kappa B kinase 偽 Ike 偽 and androgen receptor (androgen receptor,AR) in prostate cancer and prostatic hyperplasia, and to provide a basis for further study on the role of I 魏 B kinase 偽 (inhibitor kappa B kinase 偽 in the pathogenesis and development of prostate cancer. Methods the expression of IKK 偽 and AR in 30 cases of benign prostatic hyperplasia and 30 cases of prostate cancer were detected by SP immunohistochemical method. To analyze the expression of the two in different tissues, the correlation between them and the grade and stage of prostate cancer. 2. The results were statistically analyzed by SPSS 17.0 software, chi-square test was used and rank correlation analysis was carried out. The test standard to P0.05 for statistical significance. Results the results of immunohistochemistry showed that the positive expression rate of the cytoplasmic AR of the cell nuclear of the glandular epithelial cells was 73.33%, which was significantly higher than that of 46.67% in the prostatic hyperplasia group, and the positive expression rate of K 偽 was 73.33% in the prostate cancer group, which was significantly higher than that in the prostatic hyperplasia group (46.67%). The positive expression rate of AR in prostate cancer group was 83.33, which was significantly higher than that in prostate hyperplasia group (56.67%). The difference was statistically significant (P0.05). The correlation of IKK 偽 and AR expression in prostate cancer group was significant (P0.05). The correlation coefficient between IKK 偽 and AR was significant (P 0.05), indicating that there was a moderate negative correlation between the expression of IKK 偽 and AR and the grade of prostate cancer. The positive expression rate of IKK 偽 in poorly differentiated prostate cancer group was 93.33, which was significantly higher than that in well-differentiated prostate cancer group (P 0.05). The positive expression rate of IKK 偽 in advanced prostate cancer group was 93.75, which was significantly higher than that in early prostate cancer group (P 0.05). The expression of IKK 偽 was significantly higher than that in early prostate cancer group (P0.05). The positive expression rate of AR in highly differentiated prostate cancer group was 100, which was significantly higher than that in poorly differentiated group (P 0.05). The positive expression rate of AR was 92.86 in the early prostate cancer group and 75.00 in the advanced prostate cancer group (P0.05). Conclusion the positive rate and intensity of the expression of IKK 偽 and AR in prostate cancer are significantly higher than those in benign prostatic hyperplasia, indicating that IKK 偽 AR is closely related to prostate cancer. Detection of IKK 偽 AR contributes to the early diagnosis of prostate cancer. 2 with the increase of malignant degree of prostate cancer, the expression of IKK 偽 and AR show an upward and downward trend respectively. IKK 偽 AR can be used as a prognostic indicator of prostate cancer. 3 IKK 偽 and AR expression. And the biological behavior of prostate cancer, Pathology is closely related, and further study of its mechanism may provide new ideas or methods for the treatment of prostate cancer.
