【摘要】：目的探討阿司匹林對血小板誘導人乳腺癌MCF-7細胞(MCF-7 human breast cancer cells)上皮間質(zhì)轉(zhuǎn)化和遷移侵襲能力的影響。方法以未經(jīng)干預的MCF-7細胞為對照組,分別用不同濃度(0.5、2.0 mmol/L)的阿司匹林、花生四烯酸激活的血小板及不同濃度阿司匹林預處理花生四烯酸激活的血小板分別干預MCF-7細胞,采用細胞劃痕及Transwell實驗檢測腫瘤細胞遷移侵襲能力的變化。Western Blot檢測腫瘤細胞上皮間質(zhì)轉(zhuǎn)化標記蛋白E-cadherin及Vimentin的表達。結(jié)果活化的血小板處理后與對照組相比MCF-7細胞遷移侵襲能力顯著增強(P0.05),Western Blot顯示E-cadherin表達顯著減少(P0.01),Vimentin的表達顯著增加(P0.01)。阿司匹林預處理血小板活化后干預MCF-7細胞,其遷移侵襲能力較活化血小板組顯著降低(P0.05),E-cadherin表達顯著增高(P0.01),Vimentin的表達顯著減少(P0.05)。阿司匹林單獨處理后的MCF-7細胞遷移及侵襲能力較對照組無顯著變化(P0.05),E-cadherin及Vimentin的表達亦無顯著改變(P0.05)。結(jié)論體外實驗結(jié)果表明,阿司匹林可通過抗血小板活化,抑制血小板誘導的乳腺癌MCF-7細胞上皮間質(zhì)轉(zhuǎn)化和遷移侵襲能力。
[Abstract]:Objective to investigate the effects of aspirin on the translocation and invasion of human breast cancer MCF-7 cells (MCF-7 human breast cancer cells) induced by platelets. Methods the untreated MCF-7 cells were treated with different concentrations of aspirin (0.5 mmol/L), arachidonic acid-activated platelets and different concentrations of aspirin pretreated with arachidonic acid-activated platelets, respectively. Cell scratch and Transwell assay were used to detect the change of migration and invasion ability of tumor cells. Western Blot was used to detect the expression of E-cadherin and Vimentin. Results compared with the control group, the migration and invasion ability of MCF-7 cells was significantly increased (P0.05). Western Blot showed that the expression of E-cadherin decreased significantly (P0.01) and the expression of Vimentin increased significantly (P0.01). After pretreatment of platelet activation with aspirin, the migration and invasion ability of MCF-7 cells was significantly lower than that of activated platelet group (P0.05), the expression of E-cadherin was significantly increased (P0.01) and the expression of Vimentin was significantly decreased (P0.05). The expression of E-cadherin and Vimentin in MCF-7 cells treated with aspirin alone had no significant change compared with the control group (P0.05). Conclusion Aspirin can inhibit platelet-induced epithelial mesenchymal transformation and migration and invasion of breast cancer MCF-7 cells by anti-platelet activation in vitro.
