【摘要】：目的檢測(cè)丙酮酸脫氫酶激酶4(pyruvate dehydrogenase kinase 4,PDK4)在結(jié)腸癌組織及癌旁黏膜組織中的表達(dá)情況,初步探討其臨床意義。方法應(yīng)用組織芯片技術(shù)結(jié)合免疫組織化學(xué)染色方法,檢測(cè)本院收治的172例結(jié)腸癌患者術(shù)后組織標(biāo)本及86例癌旁腸黏膜組織中PDK4蛋白的表達(dá),對(duì)結(jié)果及臨床病理學(xué)參數(shù)進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果 PDK4在172例結(jié)腸癌組織中高表達(dá)率顯著高于86例癌旁腸黏膜組織(57.0%vs 26.7%,P0.01),PDK4蛋白表達(dá)高低與結(jié)腸癌的分化程度、臨床分期、淋巴結(jié)轉(zhuǎn)移、預(yù)后等密切相關(guān)(P0.05),且PDK4高表達(dá)組患者的生存時(shí)間顯著低于低表達(dá)組患者(P0.01)。在臨床分期低的結(jié)腸癌患者中,PDK高表達(dá)者預(yù)后較差(P0.01);而在臨床分期高的結(jié)腸癌患者中,PDK表達(dá)高低預(yù)后差異無統(tǒng)計(jì)學(xué)意義。結(jié)論 PDK4蛋白在結(jié)腸癌組織中表達(dá)高于癌旁腸黏膜組織,其可能參與結(jié)腸癌的發(fā)生、發(fā)展。
[Abstract]:Objective to detect the expression of pyruvate dehydrogenase kinase 4 (PDK4) in colon cancer tissues and adjacent mucosa, and to explore its clinical significance. Methods tissue microarray technique and immunohistochemical staining were used to detect the expression of PDK4 protein in the tissues of 172 patients with colon cancer and 86 patients with adjacent intestinal mucosa. The results and clinicopathological parameters were analyzed statistically. Results the high expression rate of PDK4 in 172 cases of colon cancer was significantly higher than that in 86 cases of adjacent intestinal mucosa (57.0%vs 26.7), and the expression of PDK4 protein was significantly higher than that in the adjacent mucosa of colon cancer (P0.01) and the degree of differentiation, clinical stage and lymph node metastasis of colon cancer. Prognosis was closely related (P0.05), and the survival time of patients with high expression of PDK4 was significantly lower than that of patients with low expression (P0.01). The prognosis of patients with high expression of PDK was poor in patients with low clinical stage (P0.01), but there was no significant difference in the prognosis of PDK expression in patients with high clinical stage (P0.01). Conclusion the expression of PDK4 protein in colon cancer tissues is higher than that in adjacent mucosa tissues, which may be involved in the occurrence and development of colon cancer.
