ARF在胃癌組織中的表達(dá)及臨床意義

【摘要】:目的:初步探討ARF(ADP-核糖基化因子)在胃癌組織中的表達(dá)及其臨床意義。方法:選取2013年12月至2014年6月經(jīng)手術(shù)切除并病理確診的原發(fā)性胃癌組織標(biāo)本及癌旁組織標(biāo)本各50例。應(yīng)用免疫組化檢測(cè)ARF在胃癌和癌旁組織中蛋白的表達(dá),初步探討其在胃癌及癌旁及組織中的蛋白表達(dá)水平差異及與胃癌患者臨床病理參數(shù)的相關(guān)性。應(yīng)用實(shí)時(shí)熒光定量PCR(RT-q PCR,Real-Time quantitative polymerase chain reaction)方法檢測(cè)胃癌組織及癌旁組織中ARF m RNA的表達(dá),初步探討其在胃癌組織及癌旁組織中的m RNA表達(dá)差異情況。結(jié)果:ARF在胃癌組織中的蛋白陽(yáng)性表達(dá)率為72%(36/50),顯著高于癌旁組織表達(dá)10%(5/50)(P0.05),且胃癌組織中的累積光密度值(IOD27707.02±17815.72)與癌旁組織中的累積光密度值(IOD12704.16±13932.79)比較有極顯著差異,ARF蛋白表達(dá)與胃癌患者的性別、年齡、部位、腫瘤大小及分化程度無(wú)明顯相關(guān),(P0.05),與胃癌浸潤(rùn)深度、淋巴結(jié)轉(zhuǎn)移及TNM分期有關(guān)(P0.05);胃癌組織中ARF的m RNA表達(dá)較癌旁組織升高(P0.05)。結(jié)論:(1)胃癌組織中ARF蛋白及m RNA水平的表達(dá)明顯高于癌旁組織,提示ARF蛋白及基因的檢測(cè)可能作為檢測(cè)胃癌有用的生物學(xué)標(biāo)志之一。(2)ARF蛋白表達(dá)與胃癌患者的性別、年齡、部位、腫瘤大小及分化程度無(wú)關(guān),與胃癌漿膜有無(wú)浸潤(rùn)、淋巴結(jié)轉(zhuǎn)移及TNM分期有關(guān),其可能在胃癌發(fā)生、發(fā)展中起重要作用,有可能成為判斷胃癌患者預(yù)后的標(biāo)志之一。
[Abstract]:Objective: to investigate the expression and clinical significance of ARF in gastric carcinoma. Methods: from December 2013 to June 2014, 50 specimens of primary gastric cancer and 50 specimens of paracancerous tissues were selected. Immunohistochemistry was used to detect the expression of ARF in gastric cancer and paracancerous tissues. The expression level of ARF in gastric cancer and adjacent tissues was preliminarily studied and the correlation between ARF and clinicopathological parameters of gastric cancer was investigated. The expression of ARF mRNA in gastric cancer tissues and adjacent tissues was detected by RT-q PCR Real-Time quantitative polymerase chain reaction) method, and the difference of ARF mRNA expression in gastric cancer tissues and paracancerous tissues was preliminarily investigated. Results the positive rate of protein expression in gastric cancer was 72% (36 / 50), which was significantly higher than that in paracancerous tissues (10%, 5 / 50) (P0.05). The cumulative optical density (IOD27707.02 鹵17815.72) in gastric cancer tissues was significantly different from that in adjacent tissues (IOD12704.16 鹵13932.79). To the gender of patients with gastric cancer, Age, location, tumor size and differentiation were not significantly related to the depth of gastric cancer invasion, lymph node metastasis and TNM staging (P0.05); the expression of ARF mRNA in gastric cancer tissue was higher than that in adjacent tissues (P0.05). Conclusion: (1) the expression of ARF protein and mRNA in gastric cancer tissues is significantly higher than that in adjacent tissues, suggesting that the detection of ARF protein and gene may be one of the useful biological markers for the detection of gastric cancer. (2) the expression of ARF protein and the sex, age and location of gastric cancer patients. Tumor size and differentiation are not related to serous infiltration, lymph node metastasis and TNM staging of gastric cancer, which may play an important role in the occurrence and development of gastric cancer, and may be one of the markers to judge the prognosis of gastric cancer patients.
