【摘要】：目的檢測細(xì)胞周期蛋白Cyclin Y在直腸癌和正常直腸組織中的表達(dá)情況,并將其與直腸癌及多個臨床病理因素進(jìn)行分析,探討Cyclin Y在直腸癌組織中的異常表達(dá)及其臨床意義。方法收集煙臺毓璜頂醫(yī)院2014年7月~2015年6月住院行直腸癌根治術(shù)的患者共72例,所有入組患者術(shù)前均未經(jīng)過手術(shù),放化療及生物治療等。均行直腸癌根治術(shù),并由同一手術(shù)團(tuán)隊完成。并在這72例直腸癌手術(shù)切除標(biāo)本中選取30例距腫瘤5cm以上的直腸黏膜組織作為正常直腸黏膜進(jìn)行對照。采用免疫組織化學(xué)法檢測Cyclin Y在30例正常直腸組織、72例直腸癌組織中的表達(dá)情況。并用卡方檢驗、Kruskal-Wallis檢驗等統(tǒng)計學(xué)方法進(jìn)行分析。結(jié)果1 Cyclin Y蛋白在直腸癌細(xì)胞中的表達(dá)定位以細(xì)胞膜和細(xì)胞質(zhì)為主,細(xì)胞核中表達(dá)陰性。2直腸癌組織中Cyclin Y表達(dá)陽性率明顯高于正常黏膜組織,差異具有統(tǒng)計學(xué)意義(χ~2=13.86,p0.05)。3不同病理分期的直腸癌患者的直腸癌組織中,Cyclin Y蛋白在的表達(dá)水平不同,差異具有統(tǒng)計學(xué)意義。(χ~2=7.955,p0.05)4直腸癌中Cyclin Y蛋白達(dá)陽性率在有淋巴結(jié)轉(zhuǎn)移患者中明顯高于無轉(zhuǎn)移患者,差異有統(tǒng)計學(xué)意義(p0.05);不同性別、年齡、家族史、是否侵及漿膜層及腫瘤大小不同的患者直腸癌組織中的表達(dá)水平差異沒有統(tǒng)計學(xué)意義(p0.05)。結(jié)論Cyclin Y在直腸癌中存在過表達(dá)現(xiàn)象,直腸癌中Cyclin Y蛋白達(dá)陽性率在有淋巴結(jié)轉(zhuǎn)移患者中明顯高于無轉(zhuǎn)移患者,不同病理分期的直腸癌患者的直腸癌組織中Cyclin Y蛋白在的表達(dá)水平不同,可能在直腸癌發(fā)生發(fā)展過程中起重要的調(diào)節(jié)作用。
[Abstract]:Objective to detect the expression of cyclin Y in rectal cancer and normal rectal tissues, and to analyze the expression of cyclin Y in rectal cancer and other clinicopathological factors, and to explore the clinical significance of cyclin Y in rectal cancer. Methods from July 2014 to June 2015, 72 patients with rectal cancer underwent radical resection in Yantai Yuzhouding Hospital. All the patients were not operated, chemoradiotherapy and biotherapy before operation. All patients underwent radical resection of rectal cancer, which was performed by the same surgical team. 30 cases of rectal mucosa above 5cm were selected as normal rectal mucosa. Immunohistochemical method was used to detect the expression of cyclin Y in 30 normal rectal tissues (n = 72) and rectal carcinoma (n = 72). Statistical methods such as chi-square test and Kruskal-Wallis test were used. Results 1Cyclin Y protein was mainly expressed in cell membrane and cytoplasm in rectal cancer cells. The positive rate of Cyclin Y expression in nuclear negative .2 rectal cancer tissues was significantly higher than that in normal mucosal tissues. The positive rate of Cyclin Y protein in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (p0.05). There was no significant difference in expression levels in rectal cancer with or without invasion of serous layer and tumor size (p0.05). Conclusion there is overexpression of Cyclin Y in rectal cancer, and the positive rate of Cyclin Y protein in rectal cancer is significantly higher than that in patients with lymph node metastasis. The expression level of Cyclin Y protein in rectal cancer tissues with different pathological stages is different, which may play an important role in the progression of rectal cancer.
