小干擾RNA沉默CXCR4和CXCR7對(duì)人子宮內(nèi)膜癌Ishikawa細(xì)胞裸鼠移植瘤生長(zhǎng)的抑制作用

【摘要】:目的:探討針對(duì)趨化因子受體基因CXCR4和CXCR7的小干擾RNA(siRNA)對(duì)人子宮內(nèi)膜癌Ishikawa細(xì)胞裸鼠移植瘤生長(zhǎng)的影響。方法:將子宮內(nèi)膜癌Ishikawa細(xì)胞株注射于裸鼠肩胛背部皮下建立動(dòng)物模型,待腫瘤最長(zhǎng)徑達(dá)5~7mm后,將所有荷瘤裸鼠隨機(jī)分組,分別給予CXCR4-siRNA和CXCR7-siRNA單獨(dú)或聯(lián)合瘤體內(nèi)注射,并以陰性對(duì)照siRNA和生理鹽水作為對(duì)照組,每3 d治療一次,共6個(gè)治療周期。觀察各組裸鼠移植瘤的生長(zhǎng),比較各組移植瘤的體積和質(zhì)量的差異,并以實(shí)時(shí)熒光定量RT-PCR、Western blotting和免疫組化技術(shù)驗(yàn)證CXCR4和CXCR7的基因沉默效果。結(jié)果:成功建立Ishikawa細(xì)胞裸鼠移植瘤模型,與陰性對(duì)照siRNA組和空白對(duì)照組相比,CXCR4 siRNA治療組、CXCR-siRNA治療組和CXCR4-siRNA+CXCR7-siRNA治療組的腫瘤的生長(zhǎng)均明顯受到抑制(均P0.05);CXCR4-siRNA和/或CXCR7-siRNA瘤體內(nèi)直接注射能顯著下調(diào)CXCR4、CXCR7 mRNA(均P0.05)和蛋白(均P0.01)的表達(dá)。結(jié)論:siRNA干擾CXCR4和CXCR7表達(dá)能夠有效抑制人子宮內(nèi)膜癌Ishikawa細(xì)胞裸鼠移植瘤的生長(zhǎng)。
[Abstract]:Aim: to investigate the effects of small interfering RNA (siRNA) targeting chemokine receptor genes CXCR4 and CXCR7 on the growth of human endometrial carcinoma cell line Ishikawa in nude mice. Methods: Ishikawa cell line of endometrial carcinoma was injected subcutaneously into the dorsal scapula of nude mice to establish an animal model. After the tumor reached the maximum diameter of 5~7mm, all the nude mice were randomly divided into two groups: CXCR4-siRNA and CXCR7-siRNA were injected separately or intraperitoneally. The control group was treated with siRNA and normal saline every 3 days for 6 cycles. The growth of xenografts in nude mice was observed, the volume and quality of transplanted tumors were compared, and the gene silencing effect of CXCR4 and CXCR7 was verified by real-time fluorescence quantitative RT-PCR Western blotting and immunohistochemical technique. Results: Ishikawa cell model was successfully established in nude mice. Compared with negative control siRNA group and blank control group, CXCR4 siRNA treatment group and CXCR4-siRNA CXCR7-siRNA group were significantly inhibited from tumor growth (P0.05) and / or CXCR7-siRNA injection could significantly down-regulate the expression of CXCR4CXCR7 mRNA (P0.05) and CXCR7-siRNA protein (P0.01). Conclusion interfering with the expression of CXCR4 and CXCR7 can effectively inhibit the growth of human endometrial carcinoma cell line Ishikawa in nude mice.
