【摘要】：目的:研究miRNA-195對(duì)U251細(xì)胞株CCND1表達(dá)的影響,試探miRNA-195與U251周期的關(guān)系,從而為臨床上總結(jié)膠質(zhì)瘤治療要領(lǐng)提供一定的基礎(chǔ)。方法:通過脂質(zhì)體Lipofectamin 2000將MiRNA-195mimics、MiRNA-195inhibitor轉(zhuǎn)染至膠質(zhì)瘤細(xì)胞U251,Real-time PCR檢測轉(zhuǎn)染前后MiRNA-195的含量以確保轉(zhuǎn)染效率;再通過Real-time PCR檢測轉(zhuǎn)染前后CCND1mRNA的表達(dá);Western Blotting檢測膠質(zhì)瘤細(xì)胞U251中CCND1的表達(dá)水平;應(yīng)用SPSS13.0統(tǒng)計(jì)軟件處理數(shù)據(jù),定量資料采用(?)±s描述,多組間數(shù)據(jù)比較采用單因素方差分析,組間兩兩比較采用LSD-t檢驗(yàn),P0.05為差異有統(tǒng)計(jì)學(xué)意義。結(jié)果:1、轉(zhuǎn)染MiRNA-195mimics的U251膠質(zhì)瘤細(xì)胞中的CCND1mRNA的表達(dá)高于轉(zhuǎn)染MiRNA-195inhibitor的U251膠質(zhì)瘤細(xì)胞;2、轉(zhuǎn)染MiRNA-195mimics的U251膠質(zhì)瘤細(xì)胞中的CCND1蛋白的表達(dá)高于轉(zhuǎn)染MiRNA-195inhibitor的U251膠質(zhì)瘤細(xì)胞。結(jié)論:1、MiRNA-195對(duì)膠質(zhì)瘤U251細(xì)胞株CCND1表達(dá)有影響;2、過表達(dá)MiRNA-195下調(diào)膠質(zhì)瘤U251細(xì)胞CCND1的表達(dá)。
[Abstract]:Objective: To study the effect of miRNA-195 on the expression of CCND1 in U251 cell line and explore the relationship between miRNA-195 and U251 cycle, so as to provide a certain basis for clinical summary of the treatment of glioma. Method: MiRNA-195mimics, MiRNA-195inhibitor is transferred to U251 in glioma cells through liposome Lipofectamin 2000, and Real-time PCR is used to detect the transfection before transfection. The transfection efficiency was ensured by the content of post MiRNA-195, and the expression of CCND1mRNA was detected before and after transfection by Real-time PCR; Western Blotting was used to detect the expression of CCND1 in glioma cell U251; the data were processed with SPSS13.0 statistical software, and the quantitative data were described with (?) + s, and the multiple groups of data were compared by single factor analysis of variance, 22 between groups. Compared with LSD-t test, the difference was statistically significant. Results: 1, the expression of CCND1mRNA in the U251 glioma cells transfected with MiRNA-195mimics was higher than that of U251 glioma cells transfected with MiRNA-195inhibitor; 2, the expression of CCND1 egg white in the U251 glioma cells transfected with MiRNA-195mimics was higher than that of MiRNA-195inhibitor U251 gum. Tumor cells. Conclusion: 1, MiRNA-195 has an effect on the expression of CCND1 in glioma cell line U251; 2, overexpression of MiRNA-195 down-regulation of CCND1 expression in glioma U251 cells.
