【摘要】：基因編輯技術(shù)能夠?qū)蚪M序列進(jìn)行準(zhǔn)確、穩(wěn)定的遺傳改造,給生命科學(xué)和臨床治療帶來(lái)革命性的變革。CRISPR/Cas9技術(shù)是基因編輯領(lǐng)域一個(gè)突破性進(jìn)展,它操作簡(jiǎn)單、編輯效率高,極大地?cái)U(kuò)展了科學(xué)家對(duì)基因的操控能力。本文中,筆者將介紹基因編輯技術(shù)的分類、CRISPR/Cas9技術(shù)的應(yīng)用(基因敲除、基因敲入、CRISPR/Cas9導(dǎo)入細(xì)胞的方法、提高CRISPR/Cas9特異性和調(diào)控CRISPR/Cas9系統(tǒng))、CRISPR/cas9系統(tǒng)的其他應(yīng)用(堿基轉(zhuǎn)換、表觀遺傳調(diào)控、內(nèi)源基因的轉(zhuǎn)錄調(diào)控及全基因組范圍內(nèi)的遺傳篩選)及CRISPR/Cas9系統(tǒng)在癌癥研究中的應(yīng)用(建立癌癥的小鼠模型、建立染色體重排的癌癥模型、高通量篩查與腫瘤細(xì)胞轉(zhuǎn)移相關(guān)的基因和癌癥治療),著重介紹其在癌癥領(lǐng)域的應(yīng)用和發(fā)展?fàn)顩r,使讀者對(duì)該技術(shù)有一個(gè)總體的把握。
[Abstract]:Gene editing technology can carry out accurate and stable genetic transformation of genome sequence, bring revolutionary change to life science and clinical treatment. CRISPRP / Cas9 technology is a breakthrough in gene editing field. It is simple to operate and highly efficient. It greatly expands the ability of scientists to manipulate genes. In this paper, the author will introduce the classification of gene editing technology and the application of CRISPR-Cas9 (gene knockout, gene knockout into CRISPR-Cas9 cells), improve the specificity of CRISPRP / Cas9 and regulate other applications of CRISPRP / Cas9 (base conversion, epigenetic regulation, CRISPRP / Cas9). Transcriptional regulation of endogenous genes and genome-wide genetic screening) and the application of the CRISPR-Cas9 system in cancer research (establishing cancer models in mice, establishing cancer models of chromosome rearrangements, High-throughput screening of genes and cancer therapy associated with tumor cell metastasis is focused on its application and development in the field of cancer, so that readers have a general grasp of the technology.
