拉帕替尼抑制HL60細(xì)胞增殖和促進(jìn)細(xì)胞凋亡的機(jī)制

【摘要】:目的探討拉帕替尼對(duì)急性髓細(xì)胞白血病HL60細(xì)胞增殖和凋亡的影響及其相關(guān)分子機(jī)制。方法用(5、10、15)μmol/L的拉帕替尼處理HL60細(xì)胞24 h,光學(xué)顯微鏡下觀察細(xì)胞形態(tài)變化,CCK-8法檢測細(xì)胞活力,集落形成實(shí)驗(yàn)檢測細(xì)胞增殖能力,異硫氰酸熒光素標(biāo)記的膜聯(lián)素Ⅴ/碘化丙啶(annexinⅤ-FITC/PI)雙標(biāo)記法結(jié)合流式細(xì)胞術(shù)檢測細(xì)胞凋亡,Wright改良染色(LIU染色)和Hoechst33342熒光染色觀察細(xì)胞核形態(tài);Western blot法檢測Bax、Bcl2、胱天蛋白酶3(caspase-3)、caspase-9、裂解型caspase-3(c-caspase-3)、裂解型caspase-9(c-caspase-9)、裂解型多腺苷二磷酸核糖聚合酶(c-PARP)、蛋白磷酸酶2A細(xì)胞增殖調(diào)節(jié)抑制因子(CIP2A)、c-MYC、AKT和磷酸化AKT(p-AKT)蛋白水平。結(jié)果與對(duì)照組相比,拉帕替尼能以劑量依賴的方式抑制HL60細(xì)胞的增殖,促進(jìn)細(xì)胞凋亡,誘導(dǎo)細(xì)胞產(chǎn)生核碎裂、染色質(zhì)凝聚。下調(diào)Bcl2蛋白的表達(dá),上調(diào)Bax、c-caspase-3、c-caspase-9和c-PARP蛋白水平,下調(diào)CIP2A、p-AKT和c-MYC蛋白水平。結(jié)論拉帕替尼能夠抑制HL60細(xì)胞增殖并促進(jìn)細(xì)胞凋亡,這一作用可能與下調(diào)CIP2A/AKT/c-MYC信號(hào)通路有關(guān)。
[Abstract]:Objective to investigate the effect of rapatinib on proliferation and apoptosis of HL60 cells and its molecular mechanism. Methods HL60 cells were treated with Lapatini (10 渭 mol / L) for 24 h. The morphological changes of HL60 cells were observed under optical microscope.
