pH響應(yīng)型近紅外菁類熒光探針的合成及細胞成像

【摘要】:癌癥的早期診治是提高癌癥患者治愈率的關(guān)鍵。但傳統(tǒng)的"always on"型顯影劑存在自身背景干擾且易造成"假陽性"等問題。本文利用腫瘤細胞具有弱酸性這一特性,設(shè)計合成了p H激活型不對稱菁類熒光探針,并選用氨基葡萄糖作為修飾基團以增強探針母體的水溶性并賦予其腫瘤靶向性。該探針具有p H調(diào)控的"off-on"可逆的近紅外熒光特性,以及與腫瘤弱酸性微環(huán)境相吻合的p H響應(yīng)范圍。此外,探針的濃度高達1.0×10~(-5)mol/L時仍未表現(xiàn)出明顯的細胞毒性。該探針在細胞水平實現(xiàn)了腫瘤細胞弱酸性微環(huán)境的特異性成像。
[Abstract]:Early diagnosis and treatment of cancer is the key to improve the cure rate of cancer patients. But the traditional "always on" type contrast agent has its own background interference and is easy to cause "false positive" and so on. In this paper, we designed and synthesized p-H-activated asymmetric cyanine fluorescent probes based on the weak acidity of tumor cells, and used glucosamine as the modified group to enhance the water solubility of the probe mother and endow it with tumor targeting. The probe has the reversible near infrared fluorescence characteristics of "off-on" regulated by pH and the pH response range consistent with the weak acidic microenvironment of the tumor. In addition, when the probe concentration was up to 1.0 脳 10 ~ (-5) mol / L, the cytotoxicity of the probe was not obvious. The probe achieves specific imaging of tumor cells in a weak acidic microenvironment at the cell level.
