【摘要】：目的:探討Rho相關(guān)卷曲螺旋形成蛋白激酶2(Rho associated coiled coil containing proteinkinase 2,Rock2)調(diào)控CCAAT增強(qiáng)子結(jié)合蛋白Delta(CCAAT/enhancer-binding protein delta,CEBPD)的表達(dá)對肝癌細(xì)胞增殖功能的影響。方法:1、用熒光定量PCR法檢測20例肝癌組織及對應(yīng)癌旁組織中Rock2和CEBPD的m RNA表達(dá),同時分析它們之間的關(guān)聯(lián);2、用Western blot法檢測20例肝癌組織及對應(yīng)癌旁組織中Rock2和CEBPD的蛋白表達(dá),同時分析它們之間的關(guān)聯(lián);3、用熒光定量PCR法觀察降低Rock2細(xì)胞中CEBPD m RNA表達(dá)變化;4、用Western blot法觀察降低Rock2細(xì)胞中CEBPD蛋白表達(dá)變化;5、采用EDU實(shí)驗(yàn)和CCK8實(shí)驗(yàn)觀察無意干擾對照組、下調(diào)表達(dá)Rock2組HCCLM3、SMMC7721肝癌細(xì)胞增殖能力的變化。結(jié)果:1、Rock2在肝癌組織表達(dá)明顯高于癌旁組織,CEBPD在肝癌組織表達(dá)明顯低于癌旁組織,且兩者的表達(dá)呈負(fù)相關(guān)性。2、沉默肝癌細(xì)胞中Rock2的表達(dá)后發(fā)現(xiàn)CEBPD的m RNA和蛋白表達(dá)水平上調(diào);3、Rock2通過調(diào)節(jié)CEBPD表達(dá)從而影響肝癌細(xì)胞的增殖功能。結(jié)論:在原發(fā)性肝癌細(xì)胞中沉默Rock2可引起CEBPD表達(dá)上調(diào),進(jìn)而抑制肝癌細(xì)胞的增殖功能。
[Abstract]:Aim: to investigate the effect of Rho associated crimp forming protein kinase (2(Rho associated coiled coil containing proteinkinase 2) on the proliferation of hepatocellular carcinoma (HCC) cells by regulating the expression of CCAAT enhancer binding protein Delta(CCAAT/enhancer-binding protein delta-CEBPD. Methods the m RNA expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were detected by fluorescence quantitative PCR, and the expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were analyzed by Western blot method. At the same time, the association between them was analyzed. The changes of CEBPD m RNA expression in Rock2 cells were observed by fluorescence quantitative PCR method. The changes of CEBPD protein expression in Rock2 cells were observed by Western blot assay. The unintentional interference control group was observed by EDU experiment and CCK8 experiment. The proliferation ability of HCCLM3 and SMMC7721 hepatoma cells was down-regulated in Rock2 group. Results the expression of Rock2 in HCC tissue was significantly higher than that in paracarcinomatous tissue (CEBPD), which was significantly lower than that in paracancerous tissue. After silencing the expression of Rock2 in hepatoma cells, it was found that the expression of m RNA and protein of CEBPD was up-regulated by regulating the expression of CEBPD, thus affecting the proliferation of HCC cells. Conclusion: silencing Rock2 can induce up-regulation of CEBPD expression and inhibit the proliferation of HCC cells.
