【摘要】：目的:探討Rho相關卷曲螺旋形成蛋白激酶2(Rho associated coiled coil containing proteinkinase 2,Rock2)調控CCAAT增強子結合蛋白Delta(CCAAT/enhancer-binding protein delta,CEBPD)的表達對肝癌細胞增殖功能的影響。方法:1、用熒光定量PCR法檢測20例肝癌組織及對應癌旁組織中Rock2和CEBPD的m RNA表達,同時分析它們之間的關聯;2、用Western blot法檢測20例肝癌組織及對應癌旁組織中Rock2和CEBPD的蛋白表達,同時分析它們之間的關聯;3、用熒光定量PCR法觀察降低Rock2細胞中CEBPD m RNA表達變化;4、用Western blot法觀察降低Rock2細胞中CEBPD蛋白表達變化;5、采用EDU實驗和CCK8實驗觀察無意干擾對照組、下調表達Rock2組HCCLM3、SMMC7721肝癌細胞增殖能力的變化。結果:1、Rock2在肝癌組織表達明顯高于癌旁組織,CEBPD在肝癌組織表達明顯低于癌旁組織,且兩者的表達呈負相關性。2、沉默肝癌細胞中Rock2的表達后發(fā)現CEBPD的m RNA和蛋白表達水平上調;3、Rock2通過調節(jié)CEBPD表達從而影響肝癌細胞的增殖功能。結論:在原發(fā)性肝癌細胞中沉默Rock2可引起CEBPD表達上調,進而抑制肝癌細胞的增殖功能。
[Abstract]:Aim: to investigate the effect of Rho associated crimp forming protein kinase (2(Rho associated coiled coil containing proteinkinase 2) on the proliferation of hepatocellular carcinoma (HCC) cells by regulating the expression of CCAAT enhancer binding protein Delta(CCAAT/enhancer-binding protein delta-CEBPD. Methods the m RNA expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were detected by fluorescence quantitative PCR, and the expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were analyzed by Western blot method. At the same time, the association between them was analyzed. The changes of CEBPD m RNA expression in Rock2 cells were observed by fluorescence quantitative PCR method. The changes of CEBPD protein expression in Rock2 cells were observed by Western blot assay. The unintentional interference control group was observed by EDU experiment and CCK8 experiment. The proliferation ability of HCCLM3 and SMMC7721 hepatoma cells was down-regulated in Rock2 group. Results the expression of Rock2 in HCC tissue was significantly higher than that in paracarcinomatous tissue (CEBPD), which was significantly lower than that in paracancerous tissue. After silencing the expression of Rock2 in hepatoma cells, it was found that the expression of m RNA and protein of CEBPD was up-regulated by regulating the expression of CEBPD, thus affecting the proliferation of HCC cells. Conclusion: silencing Rock2 can induce up-regulation of CEBPD expression and inhibit the proliferation of HCC cells.
