環(huán)孢素A對(duì)人胃癌BGC823細(xì)胞增殖與凋亡的影響

【摘要】:目的研究環(huán)孢素A(Cs A)對(duì)人胃癌BGC823細(xì)胞的生長(zhǎng)增殖以及凋亡的影響。方法 MTT法檢測(cè)不同濃度Cs A處理24、48、72 h后對(duì)BGC823細(xì)胞增殖抑制率;流式細(xì)胞術(shù)(FCM)檢測(cè)細(xì)胞周期、活性氧(ROS)含量及細(xì)胞線粒體膜電位(△Ψm)改變;Annexin-V-FITC/PI雙染法檢測(cè)細(xì)胞凋亡率。結(jié)果 Cs A在5~20μmol/L濃度范圍內(nèi)和24~72 h時(shí)間范圍對(duì)BGC823細(xì)胞增殖有顯著抑制作用,與藥物劑量、作用時(shí)間呈現(xiàn)量效和時(shí)效的依賴性;FCM結(jié)果顯示Cs A濃度依耐性使細(xì)胞周期阻滯于G0/G1期,與對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(F=33.45,P0.05,P0.01);細(xì)胞凋亡率隨Cs A作用濃度增加而增加;5~20μmol/L Cs A濃度范圍細(xì)胞內(nèi)ROS含量顯著增加(F=46.17,P0.01),細(xì)胞線粒體膜電位明顯降低。結(jié)論 Cs A對(duì)BGC823細(xì)胞有抑制增殖和誘導(dǎo)凋亡作用,其機(jī)制可能與阻滯細(xì)胞生長(zhǎng)周期、細(xì)胞內(nèi)ROS含量增加以及線粒體膜電位下降相關(guān)。
[Abstract]:Objective to study the effects of cyclosporine (A(Cs A) on the growth, proliferation and apoptosis of human gastric cancer BGC823 cells. Methods MTT assay was used to detect the proliferation inhibition rate of BGC823 cells treated with different concentrations of Cs A for 72 h, and flow cytometry was used to detect cell cycle, reactive oxygen species (Ros) content and mitochondrial membrane potential (蠄 m). The apoptosis rate was detected by Annexin-V-FITC / Pi double staining. Results Cs A significantly inhibited the proliferation of BGC823 cells in the concentration range of 5 渭 mol/L (20 渭 mol/L) and 24 h for 72 h. The results showed that the concentration of Cs A was dependent on dose, time and time of action. The results showed that the cell cycle was blocked in the G0/G1 phase by the concentration of CSA in accordance with tolerance. Compared with the control group, the cell apoptosis rate increased with the increase of Cs A concentration, and the ROS content increased significantly in the range of 20 渭 mol/L Cs A concentration, and the cell mitochondrial membrane potential was significantly lower than that in the control group (P < 0.05), and the cell apoptosis rate was significantly higher than that in the control group (P < 0.05), and the cell apoptosis rate increased with the increase of the concentration of Cs A, and the cell ROS content increased significantly in the range of 20 渭 mol/L Cs A concentration. Conclusion Cs A can inhibit the proliferation and induce apoptosis of BGC823 cells, and its mechanism may be related to the arrest of cell cycle, the increase of ROS content and the decrease of mitochondrial membrane potential.
