雙靶向HGFK1溶瘤腺病毒對裸鼠人胃癌移植瘤抑制作用的研究

【摘要】:目的研究雙靶向肝細胞生長因子第一環(huán)狀結構域基因(HGFK1)溶瘤腺病毒對裸鼠人胃癌移植瘤的抑制殺傷作用。方法向裸鼠右側前腋下注射人胃癌細胞SGC7901,建立人胃癌裸鼠移植瘤模型。并分別瘤內注射雙靶向HGFK1溶瘤腺病毒(TH-AD-HGFK1),雙靶向溶瘤腺病毒(TH-AD),野生型溶瘤腺病毒(AD5)和PBS,觀察移植瘤的生長情況和裸鼠胃癌模型的生存時間。結果成功建立裸鼠胃癌移植瘤模型;試驗后28天試驗組(TH-AD-HGFK1)的裸鼠移植瘤體積為1563.61±294.47mm3和,TH-AD組為2674.55±408.92mm3,AD5組為2792.94±533.57mm3,PBS組為3372.22±421.26mm3,試驗組移植瘤體積與各組相比的P值分別為P=0.029,P=0.020,P=0.004,差異有統(tǒng)計學意義(P0.05);處死裸鼠剝離移植瘤后稱重發(fā)現(xiàn)試驗組移植瘤重量為3.31±0.44g,TH-AD組為3.59±0.34g,AD5組為3.66±0.41g,PBS組為3.91±0.48g,試驗組瘤重各組相比的P值分別為P=0.239,P=0.140,P=0.013,試驗組瘤重與PBS組相比明顯減輕(P0.05),其余各組之間則無明顯差異(P0.05)。觀察至試驗后78天試驗組裸鼠全部存活(100%),TH-AD組和AD5組裸鼠存活率均為60%,PBS組裸鼠全部死亡(0%)。結論雙靶向HGFK1溶瘤腺病毒對裸鼠人胃癌移植瘤具有抑制殺傷作用,并可延長荷瘤裸鼠的生存時間。
[Abstract]:Objective to study the inhibitory effect of double targeting hepatocyte growth factor (HGFK1) oncogene HGFK1 on xenografts of human gastric cancer in nude mice. Methods Human gastric cancer cell line SGC7901 was injected into the right anterior axillary of nude mice to establish transplanted tumor model of human gastric cancer. TH-AD-HGFK1, TH-AD5 and AD5 were injected into the tumor respectively to observe the growth of transplanted tumor and the survival time of gastric cancer model in nude mice. Results the transplanted tumor model of gastric cancer was successfully established in nude mice. 28 days after the trial, the volume of transplanted tumor of nude mice in test group was 1563.61 鹵294.47mm3 and 2674.55 鹵408.92 mm3AD5 in TH-AD group was 3372.22 鹵421.26mm3 in TH-AD group and 2792.94 鹵533.57mm3m3pBS group in TH-AD group. The volume of transplanted tumor in test group was 3372.22 鹵421.26mm3 compared with each group (P = 0.029, P = 0.020, P = 0.004, respectively), the difference was statistically significant (P 0.05). It was found that the weight of transplanted tumor was 3.31 鹵0.44 g / g TH-AD group (3.59 鹵0.34 g / d), 3.66 鹵0.41 g / g PBS group (3.91 鹵0.48 g / d), respectively (P < 0.05). The tumor weight of the test group was 0.239g / g ~ 0.140g / g TH-AD group (P < 0.05), but there was no significant difference between the other groups and the PBS group (P < 0.05). It was observed that the survival rate of all the nude mice in the experimental group was 60% and that in the AD5 group was 60%. The survival rate of all the nude mice in the test group was 100% and 100% respectively, and the survival rate of all the nude mice in the AD5 group was 100% (P < 0.05). Conclusion double targeting HGFK1 adenovirus can inhibit the growth of human gastric cancer transplanted tumor in nude mice and prolong the survival time of nude mice.
