miR-338-3p在肝癌組織中的臨床意義和對肝癌細(xì)胞的影響

【摘要】:目的:探討肝癌組織中mi RNA-338-3p基因表達(dá)的臨床意義和該基因?qū)Ω伟┘?xì)胞增殖、遷移、侵襲、上皮-間質(zhì)轉(zhuǎn)化的影響。方法:使用熒光實時定量PCR技術(shù)檢測35對肝細(xì)胞癌和癌旁組織內(nèi)mi RNA-338-3p基因表達(dá)情況,通過對比患者的一般臨床資料,分析該基因的表達(dá)意義。通過攜帶目的基因的慢病毒感染體外培養(yǎng)的肝癌細(xì)胞HCCLM3和MHCC97-H,過表達(dá)肝癌細(xì)胞中的mi RNA-338-3p。再使用MTT實驗、劃痕實驗、transwell實驗、Western Bolt實驗檢測過表達(dá)基因?qū)Ω伟┘?xì)胞的增殖、遷移、侵襲能力和上皮-間質(zhì)轉(zhuǎn)化相關(guān)蛋白的影響。結(jié)果:肝細(xì)胞癌組織中mi RNA-338-3p表達(dá)水平低于癌旁組織,并與患者腫瘤分化程度、癌栓、腫瘤直徑、血管侵犯和術(shù)后生存期具有相關(guān)性(p0.05)。MTT實驗:培養(yǎng)72小時后,實驗組肝癌細(xì)胞HCCLM3和MHCC97-H的吸光度(1.153±0.232,1.247±0.247)低于陰性對照組(1.783±0.189,1.907±0.218)。劃痕實驗:HCCLM3和MHCC97-H細(xì)胞的實驗組遷移距離分別為(89.7±30.7)μm和(164.7±33.5)μm,低于陰性對照組的(189.0±31.0)μm和(267.0±28.6)μm。Transwell實驗:HCCLM3和MHCC97-H的實驗組穿過膜細(xì)胞數(shù)為(35.3±12.2)個和(74.7±10.6)個,低于陰性對照組的(70.0±17.1)個和(120.7±22.6)個。Western Blot實驗:空白對照組和陰性對照組的各蛋白表達(dá)無差異(p0.05),實驗組的E-cad表達(dá)水平高于空白對照組和陰性對照組,而Vim表達(dá)水平低于另外兩組。結(jié)論:肝癌組織內(nèi)mi RNA-338-3p較癌旁組織呈低表達(dá)狀態(tài),低表達(dá)的mi RNA-338-3p患者較高表達(dá)者,預(yù)后更差、生存期更短。體外實驗提示,高表達(dá)mi RNA-338-3p能抑制肝癌細(xì)胞的增殖能力,并可能通過上皮-間質(zhì)轉(zhuǎn)化的過程降低肝癌細(xì)胞遷移侵襲能力。
[Abstract]:Aim: to investigate the clinical significance of mi RNA-338-3p gene expression in hepatocellular carcinoma (HCC) and its effect on proliferation, migration, invasion and epithelial-interstitial transformation of HCC cells. Methods: the expression of mi RNA-338-3p gene in hepatocellular carcinoma and paracancerous tissues was detected by fluorescence real-time quantitative PCR. The significance of the gene expression was analyzed by comparing the clinical data of the patients. Human hepatoma cells HCCLM3 and MHCC97-H were infected by lentivirus carrying the target gene, which overexpressed mi RNA-338-3p. The effects of overexpression genes on proliferation, migration, invasion and epithelial-mesenchymal transformation of hepatoma cells were detected by MTT assay and Western Bolt assay. Results: the expression of mi RNA-338-3p in hepatocellular carcinoma was lower than that in adjacent tissues, and correlated with tumor differentiation, tumor thrombus, tumor diameter, vascular invasion and survival time. The absorbance of HCCLM3 and MHCC97-H in experimental group was 1.153 鹵0.232 鹵0.247 鹵0.247), which was lower than that in negative control group (1.783 鹵0.189). The migration distances of the two groups were 89.7 鹵30.7 渭 m and 164.7 鹵33.5 渭 m, respectively, which were lower than those of the negative control group (189.0 鹵31.0) 渭 m and 267.0 鹵28.6 渭 m.Transwell. The number of cells passing through the membrane of the two groups were 35.3 鹵12.2 and 74.7 鹵10.6, respectively. Western Blot test showed that there was no difference in protein expression between the blank control group and the negative control group. The expression level of E-cad in the experimental group was higher than that in the blank control group and the negative control group, but the Vim expression level was lower than that in the other two groups. Conclusion: the expression of mi RNA-338-3p in HCC is lower than that in paracancerous tissues. In patients with low expression of mi RNA-338-3p, the prognosis is worse and the survival time is shorter. The results showed that the high expression of mi RNA-338-3p could inhibit the proliferation of hepatoma cells and decrease the ability of migration and invasion of hepatoma cells through epithelial-interstitial transformation.
