姜黃素通過影響NF-κB信號(hào)通路促進(jìn)肺癌細(xì)胞的放射敏感性

【摘要】:目的探討姜黃素聯(lián)合放射治療對(duì)肺癌NCI-H460細(xì)胞的活性及肺癌小鼠放射敏感性的影響。方法把肺癌NCI-H460細(xì)胞分為4組:空白對(duì)照組、姜黃素組、γ射線組及γ射線照射與姜黃素的聯(lián)合組;然后對(duì)各組分別應(yīng)用噻唑藍(lán)(MTT)檢測(cè)細(xì)胞增殖,Annexin-V/PI染色法檢測(cè)細(xì)胞周期分布及凋亡,Western blot檢測(cè)凋亡相關(guān)蛋白Bcl-2及Bax的表達(dá),RT-PCR檢測(cè)核因子κB(NF-κB)基因表達(dá)情況。另外,建立小鼠肺癌模型同樣分為對(duì)應(yīng)的4組,姜黃素按照1mg/kg經(jīng)尾靜脈注射于小鼠,腫瘤局部進(jìn)行劑量為5Gy射線照射,處理28d后,比較不同處理組小鼠瘤體體積。結(jié)果與空白對(duì)照組、姜黃素組和γ射線組比較,聯(lián)合組NCI-H460增殖率明顯降低(P0.05),凋亡率明顯增加(P0.05),抑凋亡蛋白Bcl-2表達(dá)明顯降低(P0.05),促凋亡蛋白Bax表達(dá)明顯增高(P0.05),NF-κB mRNA表達(dá)明顯降低(P0.05),并且腫瘤的體積有明顯減少(P0.05)。結(jié)論姜黃素可通過抑制γ射線處理?xiàng)l件下NF-κB的表達(dá)并促進(jìn)細(xì)胞的G2/M期阻滯來增強(qiáng)肺癌NCI-H460細(xì)胞對(duì)γ射線的敏感性。
[Abstract]:Objective to investigate the effects of curcumin combined with radiotherapy on the activity of lung cancer NCI-H460 cells and radiosensitivity of lung cancer mice. Methods Lung cancer NCI-H460 cells were divided into four groups: blank control group, curcumin group, 緯 -ray group and combined group of 緯 -irradiation and curcumin. The cell cycle distribution was detected by Annexin-V / Pi staining and the expression of apoptosis-related protein Bcl-2 and Bax were detected by Western blot. The expression of nuclear factor- 魏 BNF- 魏 B gene was detected by reverse transcription-polymerase chain reaction (RT-PCR). In addition, mice lung cancer model was also divided into four corresponding groups. Curcumin was injected into mice via tail vein according to 1mg/kg. Tumor local dose was 5Gy irradiation. After 28 days of treatment, the tumor volume of different treatment groups was compared. Results compared with control group, curcumin group and 緯 -ray group, In the combined group, the proliferation rate of NCI-H460 was significantly decreased, the apoptosis rate was significantly increased, the expression of anti-apoptotic protein Bcl-2 was significantly decreased, the expression of pro-apoptotic protein Bax was significantly increased, the expression of NF- 魏 B mRNA was significantly decreased, and the volume of tumor was significantly decreased. Conclusion curcumin can enhance the sensitivity of lung cancer NCI-H460 cells to 緯 -rays by inhibiting the expression of NF- 魏 B and promoting the G _ 2 / M phase arrest of the cells.
