人結(jié)直腸癌組織中乙酰膽堿相關(guān)受體α9nAChR的表達(dá)及臨床意義

【摘要】:目的:結(jié)直腸癌(colorectal cancer,CRC)是消化系統(tǒng)最常見的惡性腫瘤之一,腫瘤轉(zhuǎn)移與復(fù)發(fā)是其主要死亡原因。α9nAChR是膽堿能受體(Acetylcholine receptors,AChRs)家族的成員之一,在某些惡性腫瘤中過量表達(dá),并與腫瘤的發(fā)生、發(fā)展、侵襲、轉(zhuǎn)移過程密切相關(guān)。但α9nAchR在結(jié)直腸癌中的意義尚不清楚。本研究以采用結(jié)直腸癌患者為研究對(duì)象,檢測(cè)結(jié)直腸癌組織中α9nAChR的表達(dá)情況,并分析其表達(dá)與腫瘤臨床病理特征的關(guān)系。方法:采用免疫組化法(S-P法)檢測(cè)結(jié)直腸癌患者手術(shù)切除標(biāo)本α9nAChR蛋白的表達(dá)情況。所取標(biāo)本均為結(jié)直腸癌原發(fā)病例,術(shù)前均經(jīng)病理診斷為腺癌,而且患者術(shù)前未接受放、化療等治療。同期入組病例46例,其中男性25例,女性21例。TNM分期:Ⅰ期7例,Ⅱ期19例,Ⅲ期20例。另取距瘤旁5cm以上的正常結(jié)腸組織做對(duì)照。標(biāo)本經(jīng)4%甲醛溶液固定。結(jié)果:1α9nAChR的表達(dá)主要定位于腫瘤細(xì)胞的細(xì)胞質(zhì)和細(xì)胞膜上,一般為棕黃色或者棕褐色,在腫瘤間質(zhì)的三級(jí)淋巴組織中也有表達(dá)。α9nAChR在正常結(jié)直腸組織中幾乎不表達(dá),而在結(jié)直腸癌組織中的總表達(dá)率為63.04%。2 60歲以下患者標(biāo)本中α9nAChR的表達(dá)陽性率為33.33%,而在60歲以及60歲以上患者標(biāo)本中α9nAChR的表達(dá)陽性率為63.64%,兩者具有顯著統(tǒng)計(jì)學(xué)差異(P0.05)。3α9nAChR的表達(dá)隨著分期的進(jìn)展而增高,各分期之間陽性表達(dá)率無明顯統(tǒng)計(jì)學(xué)差異,其表達(dá)與T分期有相關(guān)性,并隨著淋巴結(jié)轉(zhuǎn)移范圍增加,α9nAChR的表達(dá)的陽性率也隨之增加。4在CEA正常的患者中,α9n AChR表達(dá)的陽性率為33.33%;在CEA增高(CEA≥5μg/L)的患者,其α9nAChR表達(dá)的陽性率為63.64%;兩者間存在顯著統(tǒng)計(jì)學(xué)差異(P0.05)。5α9nAChR的表達(dá)水平與結(jié)直腸癌患者的性別、腫瘤部位、血小板水平均無關(guān)。結(jié)論1α9nAChR在結(jié)直腸癌組織及淋巴組織中表達(dá)陽性。2α9nAChR表達(dá)與結(jié)直腸癌患者的年齡、浸潤(rùn)深度、淋巴結(jié)轉(zhuǎn)移以及CEA水平相關(guān),可能參與了腫瘤的進(jìn)展。
[Abstract]:Objective: colorectal cancer CRC is one of the most common malignant tumors in the digestive system. Tumor metastasis and recurrence are the main causes of death. 偽 9nAChR is a member of the cholinergic receptor acetylcholine receptor AChRs family, which is overexpressed in some malignant tumors. It is closely related to the occurrence, development, invasion and metastasis of tumor. However, the significance of 偽-9nAchR in colorectal cancer is unclear. In this study, the expression of 偽 9nAChR in colorectal cancer was detected and the relationship between the expression of 偽 9nAChR and clinicopathological features of colorectal cancer was analyzed. Methods: the expression of 偽 9nAChR protein in surgical specimens of colorectal cancer patients was detected by immunohistochemical staining (S-P method). All the specimens were primary colorectal cancer, which were diagnosed as adenocarcinoma by pathology before operation, and the patients were not treated with radiotherapy and chemotherapy before operation.
