Sam68基因過(guò)表達(dá)可促進(jìn)乳腺癌MCF-7細(xì)胞發(fā)生上皮-間質(zhì)轉(zhuǎn)化

【摘要】:目的 :探討Sam68(Src-associated in mitosis 68 k D)對(duì)乳腺癌MCF-7細(xì)胞上皮-間質(zhì)轉(zhuǎn)化(epithelial-mesenchymal transition,EMT)、增殖、遷移和侵襲的影響。方法 :將重組質(zhì)粒pc DNA-3-Sam68轉(zhuǎn)染至乳腺癌MCF-7細(xì)胞后,應(yīng)用實(shí)時(shí)熒光定量PCR法和蛋白質(zhì)印跡法分別檢測(cè)Sam68以及EMT標(biāo)志物E-鈣黏蛋白(E-cadherin)、N-鈣黏蛋白(N-cadherin)和波形蛋白(vimentin)m RNA及蛋白的表達(dá),CCK-8法和Transwell小室法分別檢測(cè)細(xì)胞的增殖、遷移和侵襲情況。結(jié)果:重組質(zhì)粒pc DNA-3-Sam68轉(zhuǎn)染MCF-7細(xì)胞48 h后,Sam68m RNA和蛋白明顯過(guò)表達(dá)(P值均0.05)。EMT標(biāo)志物E-cadherin、N-cadherin和vimentin m RNA的表達(dá)水平無(wú)明顯變化(P值均0.05),E-cadherin蛋白的表達(dá)水平明顯下調(diào)(P0.05),而N-cadherin和vimentin蛋白的表達(dá)水平則明顯上調(diào)(P值均0.05)。并且,細(xì)胞的增殖、遷移和侵襲能力明顯增強(qiáng)(P值均0.05)。結(jié)論 :過(guò)表達(dá)Sam68基因可增強(qiáng)乳腺癌MCF-7細(xì)胞的增殖、遷移和侵襲能力,這一作用可能與調(diào)控EMT的發(fā)生有關(guān)。
[Abstract]:Aim: to investigate the effects of Sam68(Src-associated in mitosis 68kD on the epithelial-mesenchymal transition, proliferation, migration and invasion of breast cancer MCF-7 cells. Methods: the recombinant plasmid PC DNA-3-Sam68 was transfected into breast cancer MCF-7 cells. The expression of Sam68, E-cadherin (E-cadherin) and vimentin (vimentin) RNA and protein were detected by real-time fluorescence quantitative PCR and Western blot. CCK-8 and Transwell chamber methods were used to detect cell proliferation, migration and invasion, respectively. Results: after transfection of recombinant plasmid PC DNA-3-Sam68 into MCF-7 cells 48 h later, the expression levels of 0.05).EMT markers E-cadherin and vimentin m RNA were significantly over-expressed in Sam68m RNA and protein. The expression levels of E-cadherin and vimentin m RNA were not significantly changed, but N-cadherin and vimentin eggs were significantly down-regulated in the expression of E-cadherin protein. The expression level of white was significantly up-regulated by 0.05 P values. In addition, the proliferation, migration and invasion of the cells increased significantly (P < 0.05). Conclusion: overexpression of Sam68 gene can enhance the proliferation, migration and invasion of breast cancer MCF-7 cells, which may be related to the regulation of EMT.
