【摘要】：目的比較鈣離子激活氯離子通道蛋白1(anoctamin 1,ANO1)和表皮生長因子受體(epidermal growth factor receptor,EGFR)蛋白在小鼠不同食管上皮組織病變中的表達,探討ANO1、EGFR蛋白表達與小鼠食管上皮病變的關系,為早期食管癌變診斷提供參考。方法應用化學致癌劑4-硝基喹啉-1-氧化物(4NQO)由飲水法建立小鼠食管癌前病變模型,HE染色光鏡下比較觀察食管正常組織、輕度、中度、重度異型增生組織及癌變組織變化,采用免疫組化法檢測小鼠食管不同病變組織中EGFR、ANO1蛋白表達情況。結果至實驗第24周末,共收集小鼠食管正常組織14份、異型增生組織23份(其中輕度異型增生7份、中度異型增生8份、重度異型增生8份)、癌變組織18份。其中ANO1和EGFR蛋白在食管輕度異型增生組織、中度異型增生組織和正常組織中的陽性率分別為0(0/7)、12.50%(1/8)、0(0/14)和14.29%(1/7)、12.50%(1/8)、7.14%(1/14),差異均無統(tǒng)計學意義(P0.05);ANO1和EGFR在癌組織中的陽性率分別為55.56%(10/18)和61.11%(11/18),與ANO1和EGFR在異型增生組織(輕度、中度和重度增生組織)中的陽性率17.39%(4/23)和26.09%(6/23)及其正常組織中的陽性率0(0/14)和7.14%(1/14)比較差異有統(tǒng)計學意義(P0.05);ANO1和EGFR在重度異型增生組織中的陽性率分別為37.50%(3/8)和50%(4/8),與正常組織比較差異有統(tǒng)計學意義(P0.05)。在18份食管癌組織中,ANO1陽性組EGFR陽性率90%(9/10),ANO1陰性組EGFR陰性率75%(6/8),兩種蛋白的表達呈正相關(r=0.663,P=0.003)。結論 ANO1、EGFR蛋白在小鼠食管癌組織及重度異型增生上皮組織中存在過表達,且兩種蛋白在食管癌組織中的表達呈正相關,提示ANO1和EGFR兩者有可能成為食管癌早期診斷的生物學標志物。
[Abstract]:Objective to compare the expression of calcium activated chloride channel protein (1(anoctamin 1) and epidermal growth factor receptor (EGFR) protein in different esophageal epithelial lesions in mice, and to explore the relationship between the expression of ANO1 + EGFR protein and mouse esophageal epithelial lesions. To provide reference for early diagnosis of esophageal carcinoma. Methods using the chemical carcinogen 4-nitroquinoline-1-oxide-4NQO to establish mouse model of esophageal precancerous lesion by drinking water, the changes of normal esophageal tissues, mild, moderate and severe dysplasia and cancerous tissues were observed under light microscope. The expression of EGFR- ANO1 protein in different tissues of mouse esophagus was detected by immunohistochemical method. Results by the end of the 24th week of the experiment, 14 normal tissues of mouse esophagus were collected, 23 of them were dysplasia (7 cases of mild dysplasia, 8 cases of moderate dysplasia, 8 cases of severe dysplasia, 18 cases of cancerous tissue). ANO1 and EGFR proteins were found in mild dysplasia of esophagus. The positive rates in moderate dysplasia tissues and normal tissues were 0 / 7 and 12.50%, respectively, and 14.2929 / 1 / 7% 12.50% 12.50% and 7.14%, respectively. There was no significant difference in the positive rates of P0.05ANO1 and EGFR in cancer tissues (55.556 / 1018) and 61.11111118% (P < 0.05) and 61.111118% (P < 0.05), respectively. In moderate and severe hyperplastic tissues, the positive rates were 17.39% 4 / 23) and 26.09% 6 / 23) and the positive rates in normal tissues were 0 / 14) and 7.14% / 14) respectively. The positive rates of ANO1 and EGFR in severe dysplasia tissues were 37.50 / 3 / 8 and 50 / 4 / 8 respectively, which were significantly higher than those in normal tissues. The difference was statistically significant (P 0.05). In 18 cases of esophageal carcinoma, the positive rate of EGFR in the positive group of ANO1 was 90% and the negative rate of EGFR was 75% / 8% in the negative group of ANO1. The expression of the two proteins was positively correlated with the expression of 0.663P (0.003%). Conclusion ANO1-EGFR protein is overexpressed in mouse esophageal carcinoma and severe dysplasia epithelium, and the expression of ANO1-EGFR protein is positively related to the expression of ANO1-EGFR protein in esophageal carcinoma. The results suggest that ANO1 and EGFR may be biomarkers for early diagnosis of esophageal carcinoma.
