肝癌組織長鏈非編碼RNA RP13-143G15.3表達臨床意義分析

【摘要】:目的近幾年,大量的長鏈非編碼RNA被發(fā)現(xiàn),通過參與表觀遺傳、轉(zhuǎn)錄和翻譯等實現(xiàn)對基因表達的調(diào)控,促進腫瘤細胞的發(fā)生發(fā)展和轉(zhuǎn)移。探究長鏈非編碼RNA RP13-143G15.3在肝癌及癌旁組織中的表達水平,分析其差異表達與臨床指標之間的相關(guān)性,為肝癌的診療提供新的標志物。方法對5例肝癌及癌旁組織進行長鏈非編碼RNA(lncRNAs)表達譜篩查,選擇具有表達差異的RP13-143G15.3擴大樣本進行驗證。收集2016-03-01-2016-06-30廣西醫(yī)科大學(xué)腫瘤醫(yī)院肝膽外科進行肝癌切除術(shù)的56例患者肝癌及癌旁組織,采用實時熒光定量PCR技術(shù)檢測RP13-143G15.3的相對表達水平,并分析其差異表達與患者臨床特征、血清學(xué)指標及免疫組化指標的相關(guān)性。結(jié)果5例肝癌組織中RP13-143G15.3表達高于癌旁組織,P=0.005。在大樣本56例肝癌組織中相對表達量為0.026 5±0.006,癌旁組織中相對表達量為0.040 7±0.025,肝癌組織中RP13-143G15.3表達明顯低于相應(yīng)癌旁組織(Z=-3.842,P0.001),與芯片篩查結(jié)果不一致。RP13-143G15.3的差異表達與患者乙肝攜帶史(χ~2=4.667,P=0.031)、門脈高壓(χ~2=5.250,P=0.022)、甲胎蛋白水平(χ~2=4.791,P=0.029)、乳酸脫氫酶(Z=-2.237,P=0.025)、谷氨酸脫氫酶(Z=-2.101,P=0.036)、纖維蛋白原(t=-2.103,P=0.040)、CA72-4(Z=-2.147,P=0.032)、免疫學(xué)指標CD34(χ~2=9.516,P=0.023)及腫瘤增殖抗原(χ~2=8.000,P=0.005)有關(guān)。結(jié)論 RP13-143G15.3在肝癌組織中表達下調(diào),其可能起到抑癌作用,有望成為肝癌新的腫瘤標志物及分子靶點,為后續(xù)功能研究及機制研究提供理論基礎(chǔ)。
[Abstract]:Objective in recent years, a large number of long chain noncoding RNA have been found to regulate gene expression by participating in epigenetics, transcription and translation, and to promote the development and metastasis of tumor cells. To explore the expression level of long chain noncoding RNA RP13-143G15.3 in hepatocellular carcinoma and its adjacent tissues, and to analyze the correlation between the differential expression and clinical indexes, and to provide a new marker for the diagnosis and treatment of liver cancer. Methods 5 cases of liver cancer and its adjacent tissues were screened for long chain noncoding RNAs lncRNAs. the expanded RP13-143G15.3 samples with different expression were selected for verification. The relative expression of RP13-143G15.3 was detected by real-time fluorescence quantitative PCR in 56 patients undergoing hepatectomy by hepatobiliary surgery, Cancer Hospital, Guangxi Medical University, 2016-03-01-2016-06-30. The differential expression and clinical features of HCC were analyzed. Correlation of serological and immunohistochemical indexes. Results the expression of RP13-143G15.3 in 5 HCC tissues was higher than that in the adjacent tissues. The relative expression of RP13-143G15.3 was not consistent with the results of microarray screening. The relative expression of RP13-143G15.3 was 0.026 鹵0.006 in 56 HCC tissues and 0.040 7 鹵0.025 in paracancerous tissues. The expression of RP13-143G15.3 in HCC tissues was significantly lower than that in the corresponding adjacent tissues.
