miR-126、miR-355及Exportin-5在肺癌中表達研究

【摘要】:目的:探討miR-126、miR-355及Exportin-5在肺癌中表達及相互之間關系。方法:收集47例肺癌患者的癌組織及癌旁組織,利用TRIzol提取總RNA,使用定時熒光定量PCR檢測基因的相對表達量。結果:肺癌組織與癌旁組織中的miR-126,Exportin-5差異具有統(tǒng)計學意義(P0.05),癌旁組織中miR-126與pri-miR-126(r=-0.31)呈負相關(P0.05),癌組織中基因表達量與臨床指標之間的關系中發(fā)現(xiàn)miR-126與MRP(r=0.43)呈正相關(P0.05),Exportin-5與TOPO(r=-0.36)、pri-miR-126與飲酒(r=-0.34)、MRP(r=-0.43)呈負相關(P0.05)。結論:miR-126、Exportin-5在肺癌組織中表達下調,基因在肺癌發(fā)生發(fā)展過程中可能通過抑制基因的表達促進肺癌的發(fā)生與發(fā)展。
[Abstract]:Objective: to investigate the expression and relationship of miR-126 miR-355 and Exportin-5 in lung cancer. Methods: the cancer tissues and adjacent tissues of 47 patients with lung cancer were collected. The total RNAs were extracted by TRIzol and the relative expression of genes was detected by timed fluorescence quantitative PCR. Results: the difference of miR-126 exportin-5 between lung cancer and paracancerous tissues was statistically significant (P 0.05), and the expression of miR-126 was negatively correlated with pri-miR-126 r-0.31 (P 0.05). A positive correlation was found between miR-126 and MRPr0.43). There was a negative correlation with the alcohol consumption (r = 0.34) and MRP (r = 0.43) (P = 0.05, P = 0.05). Conclusion the expression of 1: miR-126 exportin-5 is down-regulated in lung cancer tissues, and the gene may promote the occurrence and development of lung cancer by inhibiting the expression of genes during the carcinogenesis and development of lung cancer.
