槲皮素PLGA-TPGS納米粒對小鼠肝癌異位實(shí)體瘤治療效果的研究

【摘要】:目的考察槲皮素PLGA-TPGS納米粒(QPTN)在體內(nèi)對荷腹水型肝癌高淋巴道轉(zhuǎn)移細(xì)胞HCa-F小鼠異位移植實(shí)體瘤的治療效果。方法建立荷HCa-F肝癌細(xì)胞小鼠模型后,隨機(jī)分為陰性對照組、空白納米粒組、5-氟尿嘧啶溶液(FS)組、槲皮素溶液(QTS)組、槲皮素PLGA納米粒(QPN)組和QPTN組。尾靜脈給藥,每2 d 1次,連續(xù)給藥20 d后處死小鼠,剝離腫瘤,稱質(zhì)量,測量腫瘤體積,根據(jù)公式計(jì)算腫瘤體積增長量和抑瘤率;行HE染色觀察腫瘤,全面評價(jià)QPTN對荷瘤小鼠的治療效果。結(jié)果小鼠體內(nèi)給藥10次后,QPTN組、QPN組、FS組的腫瘤體積增長量與陰性對照組相比明顯減小(P0.05或P0.01),QPTN組抑瘤率(59.07%)明顯高于QTS組(23.94%)、FS組(35.14%)和QPN組(46.14%)。HE染色結(jié)果也顯示QPTN組對小鼠腫瘤的治療效果最明顯。結(jié)論與QPN、QTS和FS相比,QPTN對荷HCa-F肝癌細(xì)胞小鼠異位實(shí)體瘤具有較好的治療效果。
[Abstract]:Objective to investigate the therapeutic effect of quercetin PLGA-TPGS nanoparticles in vivo on xenografted solid tumor in HCa-F mice with high lymphatic metastasis cells of ascites hepatoma. Methods mice bearing HCa-F hepatoma cells were randomly divided into three groups: negative control group, blank nanoparticles group, quercetin solution group, quercetin PLGA nanoparticles group and QPTN group. The mice were sacrificed every 2 days, 20 days later, the tumor was stripped, the tumor was weighed, the tumor volume was measured, the tumor volume growth and tumor inhibition rate were calculated according to the formula, and the tumor was observed by HE staining. To evaluate the therapeutic effect of QPTN on tumor-bearing mice. Results after 10 times of administration, the tumor volume increase in the FS group in the QPTN group was significantly lower than that in the negative control group (P 0.05 or P 0.01). The tumor inhibition rate in the QPTN group was significantly higher than that in the QTS group (P < 0.05) and in the QPN group (46.14.HE staining). The therapeutic effect of tumor is the most obvious. Conclusion compared with QPN- QTS and FS, QPTN has a better therapeutic effect on heterotopic solid tumor in mice bearing HCa-F hepatoma cells.
