【摘要】：目的探索人參皂苷Rg5對人食管癌細(xì)胞Eca-109的作用及機(jī)制。方法利用不同濃度人參皂苷Rg5在體外對人食管癌Eca109細(xì)胞作用不同時間,采用CCK-8法檢測細(xì)胞抑制率,Annexin V-FITC/PI雙染色后流式細(xì)胞儀檢測細(xì)胞凋亡率,PI單染法檢測細(xì)胞周期。結(jié)果在濃度分別為50、100μmol/L的人參皂苷Rg5作用人食管癌細(xì)胞24h后出現(xiàn)明顯形態(tài)學(xué)變化,同時處于S期及G2/M期細(xì)胞較對照組明顯增多,分別為53.08%、25.84%和58.44%、31.48%,流式細(xì)胞儀檢測早期凋亡率分別為11.45%、30.13%。結(jié)論人參皂苷Rg5能抑制食管癌Eca-109細(xì)胞增殖,其機(jī)制可能為增加細(xì)胞早期凋亡,引起細(xì)胞周期滯留。
[Abstract]:Objective to investigate the effect and mechanism of ginsenoside Rg5 on human esophageal carcinoma cell line Eca-109. Methods different concentrations of ginsenoside Rg5 were used to treat human esophageal carcinoma Eca109 cells for different time in vitro. The inhibition rate of human esophageal carcinoma Eca109 cells was detected by CCK-8 assay and the apoptosis rate was detected by flow cytometry and the cell cycle was detected by Pi single staining after Annexin V-FITC/PI double staining. Results after treated with ginsenoside Rg5 at the concentration of 50100 渭 mol/L for 24 hours, there were obvious morphological changes in human esophageal carcinoma cells, and the number of cells in S phase and G 2 / M phase was significantly higher than that in control group (53.08% and 58.4444%, 31.48%, respectively). The early apoptotic rate was 11.45% by flow cytometry. Conclusion ginsenoside Rg5 can inhibit the proliferation of esophageal carcinoma Eca-109 cells by increasing early apoptosis and cell cycle retention.
