MiR-205抑制ASPP2表達(dá)對宮頸癌的轉(zhuǎn)移及化療敏感性的影響

【摘要】:為進(jìn)一步了解MiR-205在宮頸癌中對上皮間質(zhì)轉(zhuǎn)化(epithelial-mesenchymal transition,EMT),以及細(xì)胞增殖和藥物敏感性的作用,通過實時熒光定量PCR(quantitative real-time polymerase chain reaction,qRT-PCR)和免疫印跡(Western blotting)對MiR-205的靶蛋白——ASPP2蛋白及EMT相關(guān)蛋白標(biāo)志物的表達(dá)進(jìn)行了分析。同時,研究通過噻唑藍(lán)(MTT)實驗和流式細(xì)胞技術(shù)探究了MiR-205對宮頸癌細(xì)胞的增殖和化療敏感性的影響。結(jié)果表明,MiR-205可以通過靶向抑制ASPP2的表達(dá),促進(jìn)宮頸癌細(xì)胞的增殖和對紫杉醇的耐藥性。研究發(fā)現(xiàn)了MiR-205/ASPP2可能是宮頸癌的治療和潛在生物標(biāo)志物。
[Abstract]:In order to further understand the role of MiR-205 in epithelial-mesenchymal transition, cell proliferation and drug sensitivity in cervical cancer,The expression of MiR-205 target protein ASPP2 and EMT related protein markers were analyzed by real-time PCR(quantitative real-time polymerase chain reactionation qRT-PCR and Western blotting.At the same time, the effects of MiR-205 on the proliferation and chemosensitivity of cervical cancer cells were investigated by MTT assay and flow cytometry.The results showed that MiR-205 could promote the proliferation and taxol resistance of cervical cancer cells by targeting the expression of ASPP2.The study found that MiR-205/ASPP2 may be a potential biomarker and treatment for cervical cancer.
