【摘要】：目的旨在探討姜黃素對(duì)口腔鱗癌細(xì)胞株HN4的增殖、侵襲和凋亡效應(yīng)及其分子機(jī)制。方法 0~60μmol/L姜黃素作用于口腔鱗癌細(xì)胞株HN4,24 h后應(yīng)用MTT法檢測(cè)細(xì)胞的生長(zhǎng)活性;劃痕實(shí)驗(yàn)檢測(cè)細(xì)胞遷移作用;Transwell實(shí)驗(yàn)測(cè)定對(duì)細(xì)胞的侵襲作用;流式細(xì)胞儀檢測(cè)細(xì)胞凋亡;Western blot測(cè)定P53、E-cadherin和Snail蛋白表達(dá)水平。結(jié)果 MTT檢測(cè)結(jié)果顯示各濃度姜黃素對(duì)癌細(xì)胞生長(zhǎng)有抑制,其中,20μmol/L姜黃素有明顯抑制。隨著姜黃素濃度的增加,對(duì)細(xì)胞凋亡有明顯作用。姜黃素處理后細(xì)胞周期生長(zhǎng)阻滯于G_0/G_1期。Snail的蛋白表達(dá)量隨著姜黃素的濃度增加而減少。P53和E-cadherin的表達(dá)量隨著姜黃素的濃度增加而增加。結(jié)論姜黃素能顯著抑制口腔鱗癌細(xì)胞株HN4的體外生長(zhǎng),上調(diào)P53蛋白表達(dá)誘導(dǎo)細(xì)胞凋亡可能是其作用機(jī)制之一。
[Abstract]:Objective to investigate the effect of curcumin on proliferation, invasion and apoptosis of oral squamous cell carcinoma cell line HN4 and its molecular mechanism.Methods #number0# 渭 mol/L curcumin was applied to oral squamous cell carcinoma cell line HN4H for 24 h. The cell growth activity was detected by MTT assay, the cell migration was detected by scratch assay and the invasion effect was determined by Transwell assay.Apoptosis was detected by flow cytometry and the expression of P 53 C-cadherin and Snail protein was detected by Western blot.Results the results of MTT showed that curcumin could inhibit the growth of cancer cells, and 20 渭 mol/L curcumin could inhibit the growth of cancer cells.With the increase of curcumin concentration, it has obvious effect on cell apoptosis.After curcumin treatment, cell cycle growth arrest at G_0/G_1 phase. Snail protein expression decreased with the increase of curcumin concentration, and the expression of. P53 and E-cadherin increased with the increase of curcumin concentration.Conclusion curcumin can significantly inhibit the growth of oral squamous cell carcinoma cell line HN4 in vitro, and upregulation of p53 protein expression may be one of its mechanisms.
