【摘要】：目的研究miR-29s家族在膠質(zhì)瘤干細(xì)胞(GSCs)中的表達(dá)水平,以及對(duì)GSCs生物學(xué)行為的影響是否存在差異。方法 8例膠質(zhì)瘤患者術(shù)中取腫瘤標(biāo)本,制備成GSCs。應(yīng)用實(shí)時(shí)PCR方法檢測(cè)miR-29a、miR-29b和miR-29c的內(nèi)源性表達(dá)。CCK-8方法檢測(cè)GSCs增殖,Transwell小室方法檢測(cè)GSCs的遷移侵襲,流式細(xì)胞儀檢測(cè)GSCs的凋亡。結(jié)果 miR-29a、miR-29b和miR-29c在GSCs中低表達(dá),外源性提高表達(dá)量后,可以抑制GSCs的增殖、遷移和侵襲能力,促進(jìn)GSCs的凋亡。結(jié)論 miR-29s家族在GSCs中起抑癌基因的作用,家族成員中以miR-29a的抑癌能力最強(qiáng)且起到主導(dǎo)作用。
[Abstract]:Objective to investigate the expression of miR-29s family in glioma stem cells (GSCs) and whether there are differences in the biological behavior of GSCs.Methods GSCs were obtained from 8 patients with glioma during operation.Real time PCR method was used to detect the endogenous expression of miR-29a miR-29b and miR-29c. CCK-8 was used to detect the proliferation of GSCs and the transwell chamber method to detect the migration and invasion of GSCs. Flow cytometry was used to detect the apoptosis of GSCs.Results miR-29a miR-29b and miR-29c were low expressed in GSCs. Exogenous increase of miR-29b and miR-29b could inhibit the proliferation, migration and invasion of GSCs and promote the apoptosis of GSCs.Conclusion miR-29s family plays the role of tumor suppressor gene in GSCs, and miR-29a is the strongest member of the family.
