激光捕獲顯微切割純化的人結(jié)腸癌組織的定量蛋白質(zhì)組學(xué)研究

【摘要】:為分析結(jié)腸上皮癌變進程中的差異表達蛋白質(zhì),篩選結(jié)腸癌診斷候選標(biāo)志物,先采用激光捕獲顯微切割技術(shù)(LCM)純化人正常結(jié)腸組織和結(jié)腸癌組織,再采用同位素標(biāo)記對蛋白質(zhì)進行相對和絕對定量(i TRAQ),然后結(jié)合強陽離子柱分離和反相液質(zhì)聯(lián)用(2D-LC-MS/MS)鑒定差異表達蛋白質(zhì)。試驗共鑒定了1 042個非冗余蛋白,篩選出與人結(jié)腸癌變相關(guān)的差異蛋白162個,其中在結(jié)腸癌中表達上調(diào)的蛋白質(zhì)68個,表達下調(diào)的蛋白質(zhì)94個,功能分析表明,這些差異蛋白質(zhì)涉及內(nèi)質(zhì)網(wǎng)蛋白質(zhì)加工、糖酵解/糖原異生、黏著斑細胞連接、調(diào)節(jié)微絲組裝和細胞外基質(zhì)受體相互作用等分子功能。進一步采用Western blotting及免疫組織化學(xué)技術(shù)(IHC)驗證了其中差異蛋白S100A9的表達,證實了定量蛋白質(zhì)組學(xué)結(jié)果的可靠性。研究表明:差異表達蛋白質(zhì)與結(jié)腸癌變相關(guān),并有可能成為結(jié)腸癌診斷的候選分子標(biāo)志物,為進一步深入研究這些蛋白質(zhì)在結(jié)腸癌發(fā)生和發(fā)展進程中的分子機制,為結(jié)腸癌的發(fā)病機制研究提供新線索。
[Abstract]:In order to analyze the differentially expressed proteins in the carcinogenesis of colon epithelium and to screen the candidate markers for the diagnosis of colon cancer, the normal colon tissue and colon cancer tissue were purified by laser capture microdissection (LCM).Then the relative and absolute quantification of the protein was carried out by isotope labeling, and then the differentially expressed protein was identified by strong cationic column separation and reversed phase liquid / mass spectrometry (RLC-MS).A total of 1,042 nonredundant proteins were identified and 162 differentially expressed proteins were screened out, including 68 up-regulated proteins and 94 down-regulated proteins in colon cancer.These differential proteins are involved in the processing of endoplasmic reticulum proteins, glycolysis / glycogen heterogeneity, adhesion of plaque cells, regulation of microfilament assembly and interaction of extracellular matrix receptors, and so on.Furthermore, Western blotting and immunohistochemical technique were used to verify the expression of differential protein S100A9, which confirmed the reliability of quantitative proteomics results.Studies have shown that differentially expressed proteins are associated with colon cancer and may become candidate molecular markers for the diagnosis of colon cancer. In order to further study the molecular mechanism of these proteins in the carcinogenesis and development of colon cancer,It provides a new clue for the study of the pathogenesis of colon cancer.
