口腔潛在惡性病變中腫瘤干細(xì)胞標(biāo)記物CD44與CD133表達(dá)研究及其與臨床因素相關(guān)性分析

【摘要】:目的:探究腫瘤干細(xì)胞標(biāo)記物CD44、CD133在口腔正常黏膜上皮、口腔潛在惡性病變和口腔鱗癌組織中的表達(dá)及相互關(guān)系,以及二者在人口腔黏膜異常增生細(xì)胞株DOK細(xì)胞株及人口腔鱗癌細(xì)胞株CAL-27細(xì)胞株中的表達(dá),同時(shí)分析CD44、CD133表達(dá)與患者臨床因素之間的關(guān)系;探究CD44和CD133在口腔潛在惡性病變發(fā)生和癌變過(guò)程中的表達(dá)規(guī)律及其可能機(jī)制,借此評(píng)價(jià)二者作為口腔潛在惡性病變癌變監(jiān)測(cè)指標(biāo)、口腔鱗癌早期檢測(cè)指標(biāo)的臨床價(jià)值。方法:采用免疫組織化學(xué)雙重染色技術(shù)檢測(cè)CD44、CD133在口腔正常黏膜、口腔扁平苔蘚、口腔白斑、口腔鱗癌組織中的表達(dá)水平。引進(jìn)并培養(yǎng)DOK細(xì)胞株和CAL-27細(xì)胞株,采用細(xì)胞免疫熒光雙重染色技術(shù)觀察兩細(xì)胞株中CD44、CD133共表達(dá)情況;采用細(xì)胞免疫化學(xué)技術(shù)檢測(cè)兩細(xì)胞株中CD44、CD133的表達(dá)含量;采用蛋白免疫印跡技術(shù)檢測(cè)兩細(xì)胞株中CD44、CD133的蛋白含量。收集青島大學(xué)醫(yī)學(xué)院附屬醫(yī)院口腔科和病理科就診并隨訪存檔的口腔黏膜潛在惡性病變及口腔鱗癌的存檔病例,分析評(píng)價(jià)患者各項(xiàng)臨床資料與CD44、CD133表達(dá)相關(guān)性。采用SPSS19.0軟件,比較口腔正常黏膜、口腔扁平苔蘚、口腔白斑、口腔鱗癌組織中CD44、CD133的表達(dá)情況。比較DOK細(xì)胞株和CAL-27細(xì)胞株中CD44、CD133的含量及變化。分析CD44、CD133表達(dá)與患者臨床因素之間的關(guān)系。探究CD44及CD133在口腔黏膜癌變過(guò)程中的表達(dá)規(guī)律及其可能機(jī)制,借此評(píng)價(jià)二者作為口腔潛在惡性病變癌變監(jiān)測(cè)指標(biāo)、口腔鱗癌早期檢測(cè)指標(biāo)的臨床價(jià)值。結(jié)果:口腔正常黏膜、口腔潛在惡性病變、口腔鱗癌三組中,CD44的表達(dá)率分別為100.00%(10/10)、96.67%(58/60)、71.67%(43/60),表達(dá)逐漸減弱(P0.05);CD133的表達(dá)率分別為0.00%(0/10)、35.00%(21/60)、63.33%(38/60),表達(dá)逐漸增強(qiáng)(P0.05);CD44與CD133的表達(dá)有一定相關(guān)性(P0.05)。DOK細(xì)胞株和CAL-27細(xì)胞株中存在CD44和CD133共表達(dá)。與DOK細(xì)胞株相比,CAL-27細(xì)胞株中CD44表達(dá)減弱,CD133表達(dá)增強(qiáng)。CD44、CD133表達(dá)與臨床因素相關(guān)性分析中,兩者表達(dá)在患者的年齡、性別、吸煙史、飲酒史及病變部位分組中的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。CD44、CD133表達(dá)在口腔鱗癌患者的各臨床分期、淋巴結(jié)轉(zhuǎn)移中的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:在口腔潛在惡性病變癌變發(fā)生和發(fā)展的過(guò)程中,CD44表達(dá)逐漸下降,CD133表達(dá)逐漸增高,且CD44和CD133兩者表達(dá)存在一定相關(guān)性,同時(shí)二者表達(dá)與口腔鱗癌的臨床分期和淋巴結(jié)轉(zhuǎn)移有關(guān)。CD44介導(dǎo)的細(xì)胞黏附與分化,CD133維持的干細(xì)胞特性可能在口腔潛在惡性病變癌變發(fā)生和發(fā)展的過(guò)程中發(fā)揮了重要作用。CD44和CD133作為口腔潛在惡性病變癌變監(jiān)測(cè)指標(biāo)、口腔鱗癌早期檢測(cè)指標(biāo)可能具有重要的臨床價(jià)值。
[Abstract]:Objective: to investigate the expression and relationship of tumor stem cell marker CD44hCD133 in normal oral mucosa epithelium, oral potential malignant lesions and oral squamous cell carcinoma.The expression of CD4pCD133 in human oral mucosal dysplasia cell line DOK cell line and human oral squamous cell carcinoma cell line CAL-27 cell line was analyzed, and the relationship between CD4pCD133 expression and clinical factors was analyzed.To explore the expression and possible mechanism of CD44 and CD133 in the process of occurrence and carcinogenesis of oral potential malignant lesions, and to evaluate their clinical value as a monitoring index for carcinogenesis of oral potential malignant lesions and early detection indexes for oral squamous cell carcinoma (OSCC).Methods: Immunohistochemical double staining technique was used to detect the expression level of CD44 and CD133 in normal oral mucosa, oral lichen planus, oral leukoplakia and oral squamous cell carcinoma.DOK cell line and CAL-27 cell line were introduced and cultured. The co-expression of CD44hCD133 in the two cell lines was observed by double immunofluorescence staining, and the expression of CD44hCD133 in the two cell lines was detected by cytoimmunochemical technique.The protein content of CD44 and CD133 in two cell lines was detected by Western blot.Cases of potential malignant lesions of oral mucosa and oral squamous cell carcinoma (SCC) were collected from Department of Stomatology and Department of Pathology of affiliated Hospital of Medical College of Qingdao University.SPSS19.0 software was used to compare the expression of CD44 and CD133 in normal oral mucosa, oral lichen planus, oral leukoplakia and oral squamous cell carcinoma.The contents and changes of CD44 and CD133 in DOK and CAL-27 cell lines were compared.To analyze the relationship between the expression of CD 44 and CD 133 and the clinical factors.To explore the expression and possible mechanism of CD44 and CD133 in the process of oral mucosa carcinogenesis, and to evaluate their clinical value as a monitoring index for the carcinogenesis of oral potential malignant lesions and early detection indexes for oral squamous cell carcinoma (OSCC).Results: normal oral mucosa, potential malignant lesions of oral cavity,鍙h厰槌炵檶涓夌粍涓,
