【摘要】：目的應(yīng)用iTRAQ標(biāo)記定量蛋白質(zhì)組學(xué)技術(shù),對正常人及大腸癌脾虛證和濕熱證患者間的唾液蛋白進行定量,并分析其差異蛋白,從而篩選出大腸癌(colorectal cancer,CRC)脾虛證、濕熱證患者唾液中的特異性蛋白質(zhì)標(biāo)志物,以期探索無創(chuàng)傷性、靈敏度高、特異性強的大腸癌病證分子診斷技術(shù)方法。方法采用同位素標(biāo)記相對和絕對定量(isobaric tags for relative and absolute quantification,iTRAQ)蛋白質(zhì)組學(xué)技術(shù),對正常人(45例)、大腸癌脾虛證(9例)及濕熱證患者(18例)的唾液進行蛋白定量;再用Excel軟件進行統(tǒng)計,將ratio≥1.5的定為上調(diào)蛋白,而ratio≤0.667的定為下調(diào)蛋白,找出其唾液中差異蛋白質(zhì);再運用Gene Ontology(GO)生物數(shù)據(jù)庫,對差異蛋白質(zhì)進行功能注釋,闡釋差異蛋白所參與的生物學(xué)過程(Biological Process,BP)、具有的分子功能(Moleculer Function,MF)以及所屬的細胞組分(Cellular Component,CC),以及對相關(guān)蛋白進行通路分析。結(jié)果1、通過對大腸癌脾虛證患者與正常人唾液對比,其中ratio≥1.5的蛋白有202個,而ratio≤0.667的蛋白有176個。2、通過對大腸癌濕熱證患者與正常人唾液對比,其中ratio≥1.5的蛋白有270個,而ratio≤0.667的蛋白有223個。3、通過對大腸癌濕熱證與脾虛證患者的唾液對比,其中ratio≥1.5,兩者相同的蛋白共有45個,濕熱證和脾虛證患者之間唾液中差異蛋白分別為157個、225個。而ratio≤0.667,相同的蛋白共有34個,濕熱證和脾虛證患者之間唾液差異蛋白分別為142個、189個。結(jié)論1、通過iTRAQ定量蛋白質(zhì)組學(xué)研究獲得大腸癌脾虛證的特征性差異蛋白質(zhì)表達譜,并篩選出本病脾虛證多個特征性蛋白質(zhì)生物標(biāo)志物;2、通過iTRAQ定量蛋白質(zhì)組學(xué)研究獲得大腸癌濕熱證的特征性差異蛋白表達譜,并篩選出本病濕熱證多個特征性蛋白質(zhì)生物標(biāo)志物;3、運用iTRAQ定量蛋白質(zhì)組學(xué)技術(shù)篩選出大腸癌脾虛證與濕熱證共有的特異性蛋白分子標(biāo)志物;4、運用生物信息技術(shù)探討特異性蛋白分子所參與的生物學(xué)過程、細胞組分和分子功能。
[Abstract]:Objective to quantify saliva protein in normal subjects and patients with spleen deficiency and damp-heat syndrome by using iTRAQ labeled quantitative proteomics technique, and to analyze the differential proteins, so as to screen out the spleen deficiency syndrome of colorectal carcinoma Colorectal carcinoma CRC.The specific protein markers in saliva of patients with dampness and heat syndrome were studied in order to explore the molecular diagnostic method of colorectal cancer syndrome without trauma, high sensitivity and strong specificity.Methods the relative and absolute quantification of isobaric tags for relative and absolute quantificationiTRAQs were used to quantify the saliva of 45 normal subjects, 9 patients with spleen deficiency syndrome of colorectal cancer and 18 patients with damp-heat syndrome.Ratio 鈮，
