【摘要】：隨著癌癥發(fā)生率和死亡率的逐年上升,癌癥已成為威脅人類(lèi)健康的首要敵人,因此越來(lái)越多的研究者投入癌癥發(fā)病機(jī)制及新療法的探索之中。盡管如此,每年仍然只有屈指可數(shù)的抗癌新藥通過(guò)審批,究其原因主要是絕大多數(shù)具有較好臨床前效應(yīng)的新藥缺乏良好的臨床療效,因此合理應(yīng)用及創(chuàng)新臨床前抗腫瘤藥物的評(píng)價(jià)體系,促進(jìn)臨床前研究與臨床藥效的結(jié)合與統(tǒng)一,就顯得尤為迫切和重要。眾所周知,動(dòng)物癌癥模型在癌癥的病因和發(fā)病機(jī)制研究以及治療評(píng)估中發(fā)揮著舉足輕重的作用。本文對(duì)最為常用的小鼠癌癥模型在臨床前研究中的發(fā)展前景以及面臨的挑戰(zhàn)進(jìn)行了探討,并對(duì)新興癌癥模型的應(yīng)用前景進(jìn)行了簡(jiǎn)述,為臨床前合理評(píng)價(jià)藥物療效提供參考。
[Abstract]:With the increasing incidence and mortality rate of cancer, cancer has become the primary threat to human health, so more and more researchers are exploring the mechanism of cancer and new therapies. Only a handful of new anticancer drugs are approved each year. The main reason is that most of the new drugs with good preclinical effects lack good clinical efficacy. Therefore, the rational application and innovation of the evaluation system of pre-clinical anti-tumor drugs, It is particularly urgent and important to promote the combination and unification of preclinical research and clinical efficacy. Animal cancer models play an important role in the study of the etiology and pathogenesis of cancer, as well as in the evaluation of treatment. In this paper, the development prospects and challenges of the most commonly used mouse cancer models in preclinical research are discussed. The application prospect of the new cancer model is briefly described, which provides a reference for the rational evaluation of the efficacy of drugs before clinical treatment.
