【摘要】：目的:探討Tg737基因過表達(dá)對(duì)人肝癌細(xì)胞株細(xì)胞周期和凋亡的影響及可能的機(jī)制。方法:將肝癌HepG2和MHCC97-H細(xì)胞分別用脂質(zhì)體法轉(zhuǎn)染pcDNA3.1-Tg737質(zhì)粒(Tg737過表達(dá)組)或pcDNA3.1空載體(空載體組),或單純脂質(zhì)體孵育(脂質(zhì)體組),以各自未處理的細(xì)胞為空白對(duì)照。48h后分別用流式細(xì)胞儀檢測(cè)細(xì)胞周期與凋亡,Hoechst33342染料法檢測(cè)細(xì)胞核形態(tài),Westernblot法檢測(cè)cyclinA、Bax、Bcl-2表達(dá)。結(jié)果:與各自的空白對(duì)照組比較,Tg737過表達(dá)組HepG2和MHCC97-H細(xì)胞的S期細(xì)胞數(shù)量與細(xì)胞凋亡率均明顯增加(均P0.05),且細(xì)胞核均出現(xiàn)明顯凋亡形態(tài)學(xué)改變,同時(shí)伴隨cyclinA、Bax的表達(dá)上調(diào)和Bcl-2的下調(diào)(均P0.05);空載體組、脂質(zhì)體組HepG2和MHCC97-H細(xì)胞鏡下未見明顯的凋亡形態(tài)學(xué)變化,且上述指標(biāo)差異均無統(tǒng)計(jì)學(xué)意義(均P0.05)。結(jié)論:Tg737基因過表達(dá)可以抑制肝癌細(xì)胞周期進(jìn)程、促進(jìn)其凋亡,機(jī)制可能與Tg737參與調(diào)節(jié)cyclinA、Bax、Bcl-2有關(guān)的信號(hào)通路有關(guān)。
[Abstract]:Objective: to investigate the effect of overexpression of Tg737 gene on cell cycle and apoptosis of human hepatoma cell line and its possible mechanism. Methods: HepG2 and MHCC97-H cells were transfected with pcDNA3.1-Tg737 plasmid Tg737 or pcDNA3.1 empty vector by liposome respectively. Empty carrier group or pure liposome incubated (liposome group). 48 hours later, flow cytometry was used to detect cell cycle and apoptosis. Hoechst33342 dye method was used to detect nuclear morphology and Western blot. Results: compared with the control group, the S phase cell number and apoptosis rate of HepG2 and MHCC97-H cells in Tg737 overexpression group were significantly increased (P 0.05), and the morphological changes of apoptosis were observed in the nuclei. At the same time, the expression of cyclin Agna Bax was up-regulated and the Bcl-2 was down-regulated (all P 0.05), but there were no obvious morphological changes of apoptosis in HepG2 and MHCC97-H cells in empty vector group, liposome group and liposome group under microscope. Conclusion the overexpression of Tg737 gene can inhibit the cell cycle progression and promote the apoptosis of hepatoma cells, and the mechanism may be related to the regulation of the signal pathway related to cyclin Agna BaxC-2 by Tg737.
