【摘要】：目的本文旨在利用Western blot法和免疫組化等技術研究受體相互作用蛋白激酶1(RIP1)蛋白在食管鱗狀細胞癌中的表達水平,并結合臨床病理資料研究其在食管鱗癌中的表達水平的相關臨床意義。方法應用Western blot法和免疫組化法檢測,分析100例食管鱗癌患者的癌組織、相應癌旁組織(距病灶邊緣2cm以內)和對應正常黏膜組織(距病灶邊緣5 cm外)中RIP1的表達水平。以正常食管粘膜組織作為陰性對照,研究RIP1在食管癌組織及對應病例癌旁組織中的差異表達,應用統(tǒng)計學進一步分析RIP1表達水平與病患年齡、性別、臨床病理分期等相關關系。結果71%食管鱗狀上皮惡性腫瘤組織中RIP1蛋白的表達水平高于對應癌旁組織和正常黏膜組織,20%食管鱗狀上皮惡性腫瘤旁組織中RIP1蛋白的表達水平高于對應食管鱗癌組織和正常食管黏膜中蛋白的表達水平,差異有統(tǒng)計學意義(P0.05)。食管鱗癌組織中高表達的RIP1蛋白水平與其TNM分期、病理分級、是否存在周圍淋巴結轉移(P0.05),與性別、年齡無關。癌旁組織中高表達的RIP1蛋白水平與腫瘤浸潤深度、TNM分期有關(P0.05),與性別、年齡、是否存在周圍淋巴結轉移及病理分級無關。結論RIP1蛋白在食管鱗癌中的高表達可能與腫瘤的病理分級、TNM分期有關,對于食管鱗癌的發(fā)生發(fā)展有一定的預測作用。
[Abstract]:Objective to investigate the expression of receptor interacting protein kinase 1 (RIP1) protein in esophageal squamous cell carcinoma by Western blot and immunohistochemistry. The clinical significance of the expression level in esophageal squamous cell carcinoma was studied with clinicopathological data. Methods Western blot method and immunohistochemical method were used to analyze the cancer tissues of 100 patients with esophageal squamous cell carcinoma. The expression of RIP1 in the corresponding paracancerous tissues (within 2cm) and the corresponding normal mucosa (5 cm away from the edge of the lesion) was detected. The normal esophageal mucosa was used as the negative control. To study the differential expression of RIP1 in esophageal carcinoma tissues and adjacent tissues of corresponding cases, and to further analyze the expression level of RIP1 with age and sex by statistical analysis. Results the expression level of RIP1 protein in 71% malignant esophageal squamous carcinoma tissues was higher than that in the corresponding paracancerous tissues and normal mucosa tissues of 20% esophageal squamous carcinoma tissues. The expression level of protein in esophageal squamous cell carcinoma tissues and normal esophageal mucosa was higher than that in corresponding esophageal squamous cell carcinoma tissues. The high expression of RIP1 protein in esophageal squamous cell carcinoma was correlated with its TNM stage, pathological grade, whether there were peripheral lymph node metastasis (P0.05), and sex. The high expression of RIP1 protein in paracancerous tissues was related to the stage of tumor invasion depth (P0.05), sex, age, and age. Conclusion the high expression of RIP1 protein in esophageal squamous cell carcinoma may be related to the pathological grade and staging of the tumor, which may predict the occurrence and development of esophageal squamous cell carcinoma.
