木鱉子單體化合物對羥基桂皮醛對食管癌移植瘤的抑制作用

【摘要】:目的:探討木鱉子單體化合物對羥基桂皮醛(p-hydroxylcinnamaldehyde,CMSP)對食管癌細(xì)胞增殖及在BALB/c裸鼠移植瘤生長的抑制作用。方法:細(xì)胞計數(shù)法檢測CMSP對食管癌細(xì)胞株Kyse 30和TE-13增殖的抑制作用,RT-PCR法檢測食管癌細(xì)胞中CEA和SCC mRNA的表達(dá)情況,Western blotting法檢測食管癌細(xì)胞中C-myc和N-myc蛋白的表達(dá)變化。構(gòu)建裸鼠食管癌細(xì)胞移植瘤模型,分為對照組和CMSP處理組,實驗結(jié)束后測量移植瘤大小及體積;免疫組織化學(xué)SP法觀察移植瘤組織C-myc和N-myc蛋白的表達(dá)變化,H-E染色后光鏡下觀察兩組裸鼠移植瘤組織和肝、脾組織的病理學(xué)變化。結(jié)果:CMSP對食管癌細(xì)胞Kyse 30和TE-13的增殖具有明顯的抑制作用[(1.7±0.3)vs(3.8±0.3),(1.6±0.2)vs(4.5±0.4),均P0.01]。經(jīng)CMSP處理后:(1)Kyse 30和TE-13細(xì)胞中CEA和SCC mRNA表達(dá)降低(P0.01)、C-myc和N-myc蛋白表達(dá)明顯低于對照組(P0.05或P0.01);(2)裸鼠移植瘤平均體積和質(zhì)量明顯降低(P0.05或P0.01),移植瘤組織中Cmyc和N-myc蛋白表達(dá)水平低于鹽水對照組腫瘤組織。CMSP處理組和對照組中肝、脾組織并未發(fā)生任何病理改變。結(jié)論:木鱉子單體化合物CMSP體外抑制食管癌細(xì)胞增殖,體內(nèi)抑制裸鼠食管癌移植瘤的生長。
[Abstract]:Objective: to investigate the inhibitory effect of monomeric compound of Trionyx chinensis on the proliferation of esophageal carcinoma cells and the growth of transplanted tumor in BALB/c nude mice. Methods: the inhibitory effect of CMSP on the proliferation of esophageal carcinoma cell lines Kyse 30 and TE-13 was detected by cell count method. The expression of CEA and SCC mRNA in esophageal carcinoma cells was detected by RT-PCR and the expression of C-myc and N-myc in esophageal carcinoma cells was detected by Western blotting. At the end of the experiment, the size and volume of transplanted tumor were measured, the expression of C-myc and N-myc protein in transplanted tumor tissues were observed by immunohistochemical SP method, and the transplanted tumor tissues and liver of two groups were observed under light microscope after H-E staining. Results the proliferation of Kyse 30 and TE-13 in esophageal carcinoma cells was significantly inhibited by CMSP treatment [1.7 鹵0.3)vs(3.8 鹵0.3 鹵0.4, respectively, P0.01]. After CMSP treatment, the expression of CEA and SCC mRNA was significantly lower than that of P0.01C-myc and N-myc. The mean volume and quality of transplanted tumor in nude mice were significantly lower than those in control group (P 0.05 or P 0.01). The expression level of Cmyc and N-myc protein in transplanted tumor tissue was lower than that in saline control group. Conclusion: CMSP can inhibit the proliferation of esophageal carcinoma cells in vitro and inhibit the growth of transplanted tumor of esophageal carcinoma in nude mice in vivo.
