【摘要】：目的觀察miR-630在胃癌組織中的表達變化,及miR-630對胃癌細胞株SGC-7901增殖、侵襲能力的影響。方法收集131例胃癌患者的胃癌組織及癌旁正常胃組織,采用實時熒光定量PCR法檢測miR-630。培養(yǎng)SGC-7901細胞,分為實驗組、空載組和對照組,實驗組轉(zhuǎn)染miR-630,空載組轉(zhuǎn)染空質(zhì)粒,對照組僅加入PBS;分別于培養(yǎng)12、24、48、72 h后采用MTT法檢測細胞增殖能力;培養(yǎng)24 h后以Transwell小室檢測細胞侵襲能力;培養(yǎng)24 h后,采用Western blotting法檢測細胞中的p27、MMP-2、MMP-9蛋白。結(jié)果胃癌組織、癌旁組織中miR-630的相對表達量分別為1.93±0.41、2.74±0.48,胃癌組織中miR-630相對表達量低于癌旁組織(P均0.05)。腫瘤直徑≥5 cm者胃癌組織中miR-630相對表達量低于直徑5 cm者,有淋巴結(jié)轉(zhuǎn)移者低于無淋巴結(jié)轉(zhuǎn)移者,浸潤深度為T2~T4級者低于T1級者,Ⅲ~Ⅳ期者低于Ⅰ~Ⅱ期者(P均0.05)。轉(zhuǎn)染24、48、72 h后,實驗組細胞增殖能力均低于轉(zhuǎn)染同時點的空載組和對照組(P均0.01)。實驗組、空載組、對照組穿膜細胞數(shù)分別為15.4±2.7、24.3±23.1、29.7±19.4,實驗組穿膜細胞數(shù)少于對照組和空載組(P均0.01)。實驗組細胞中p27蛋白相對表達量高于空載組和對照組,MMP-2、MMP-9蛋白相對表達量低于空載組和對照組(P均0.01)。結(jié)論胃癌組織中miR-630表達減少,mir-630低表達可能與胃癌的發(fā)生發(fā)展有關(guān);miR-630有抑制胃癌細胞增殖、侵襲的作用,機制可能與p27蛋白表達上調(diào)及MMP-2、MMP-9蛋白表達下調(diào)有關(guān)。
[Abstract]:Objective to observe the expression of miR-630 in gastric carcinoma and the effect of miR-630 on the proliferation and invasion of gastric cancer cell line SGC-7901. Methods gastric cancer tissues and adjacent normal gastric tissues were collected from 131 patients with gastric cancer. The cultured SGC-7901 cells were divided into experimental group, no-load group and control group. The experimental group was transfected with miR-630, the no-load group was transfected with empty plasmid, and the control group was only added into the control group. After 24 h culture, the invasion ability of the cells was detected by Transwell chamber, and the p27 MMP 2 MMP-9 protein was detected by Western blotting assay after 24 h culture. The relative expression of miR-630 in paracancerous tissues was 1.93 鹵0.41t2.74 鹵0.48.The relative expression of miR-630 in gastric cancer was lower than that in paracancerous tissues (P < 0.05).
