Rad9、Rad1、Hus1基因在結(jié)直腸癌中的表達(dá)及意義

【摘要】:目的:檢測(cè)Rad9、Rad1、Hus1基因的mRNA在結(jié)直腸癌組織及癌旁正常組織中的表達(dá)水平,并結(jié)合腫瘤的病理分化程度、分期、淋巴結(jié)轉(zhuǎn)移等臨床病理資料,探討其表達(dá)水平是否與結(jié)腸癌發(fā)病、侵襲及轉(zhuǎn)移有關(guān),為其能否成為結(jié)直腸癌重要的生物學(xué)標(biāo)志物提供參考,為直腸癌的早期診斷、早期治療、預(yù)后判斷提供依據(jù)。方法:收集35例結(jié)直腸癌組織及癌旁正常組織,采用熒光定量PCR技術(shù)檢測(cè)組織中Rad9、Rad1、Hus1基因的mRNA表達(dá)水平,分析其表達(dá)水平在癌組織及癌旁正常組織中有無(wú)差異,并進(jìn)一步分析其表達(dá)差異與腫瘤的TNM分期、病理分化程度、部位、性別、年齡等臨床病理資料是否有關(guān)。研究結(jié)果采用Spss17.0軟件進(jìn)行統(tǒng)計(jì)學(xué)分析,P0.05為差異有統(tǒng)計(jì)學(xué)意義。結(jié)果:1.25例(71.43%)結(jié)直腸癌組織中Rad9 m RNA表達(dá)量較癌旁正常組織降低,結(jié)直腸癌組織中Rad9 mRNA的表達(dá)量(2.98±5.14)低于癌旁正常組織(7.11±16.82),差異有統(tǒng)計(jì)學(xué)意義(Z=-2.064,P=0.039)。2.Rad9 mRNA在低分化腫瘤中有13例(92.85%)低表達(dá),在中分化腫瘤中有9例(64.29%)低表達(dá),在高分化腫瘤中有3例(42.86%)低表達(dá),差異有統(tǒng)計(jì)學(xué)意義(P=0.033)。3.Rad9mRNA在有淋巴結(jié)轉(zhuǎn)移的組織中有16例(88.89%)低表達(dá),無(wú)淋巴結(jié)轉(zhuǎn)移的組織中有9例(52.94%)低表達(dá),差異有統(tǒng)計(jì)學(xué)意義(P=0.027)。4.Rad9 mRNA在結(jié)直腸癌TNM分期Ⅰ期、Ⅱ期、Ⅲ期、Ⅳ期中分別有1例(25.00%)、8例(61.54%)、10例(83.33%)、6例(100%)低表達(dá),差異有統(tǒng)計(jì)學(xué)意義(P=0.042)。5.Rad9mRNA在不同年齡、性別和部位的表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。6.16例(45.71%)結(jié)直腸癌組織中Rad1mRNA的表達(dá)量較正常組織降低,結(jié)直腸癌組織中的表達(dá)量(2.01±4.62)與正常組織中的表達(dá)量(2.61±6.49)差異無(wú)統(tǒng)計(jì)學(xué)意義(Z=-0.402,P=0.712)。7.14例(40.00%)結(jié)直腸癌組織中Hus1 mRNA表達(dá)量較正常組織降低,結(jié)直腸癌中的表達(dá)量(4.04±7.51)與正常組織中的表達(dá)量(3.36±8.30)差異無(wú)統(tǒng)計(jì)學(xué)意義(Z=-0.292,P=0.784)。結(jié)論:1.結(jié)直腸癌腫瘤組織中Rad9 mRNA的表達(dá)量較癌旁正常組織降低,提示Rad9基因可能和結(jié)直腸癌的發(fā)病有關(guān)。2.結(jié)直腸癌組織中Rad9 mRNA的低表達(dá)率隨淋巴結(jié)轉(zhuǎn)移、病理分化程度降低及腫瘤的分期級(jí)別增高而升高,提示Rad9基因可能與結(jié)直腸癌的侵襲及轉(zhuǎn)移有關(guān)。3.目前尚不能認(rèn)為Rad9基因的表達(dá)水平與年齡、性別及腫瘤部位有關(guān)。4.目前尚不能認(rèn)為Rad1、Hus1基因與結(jié)直癌發(fā)病有關(guān)。
[Abstract]:Objective: to detect the expression level of Rad9- Rad1hHus1 gene in colorectal cancer and adjacent normal tissues, and to explore whether the expression level of Rad9- Rad1hHus1 gene is associated with the incidence of colon cancer, in combination with the clinical pathological data, such as pathological differentiation, staging, lymph node metastasis, and so on. Invasion and metastasis can be used as an important biomarker for colorectal cancer. Methods: 35 cases of colorectal cancer tissues and adjacent normal tissues were collected for early diagnosis, early treatment and prognosis judgment. Fluorescence quantitative PCR technique was used to detect the expression of Rad9hRd1hHus1 gene in tissues, and to analyze whether there were differences in the expression levels of Rad9hRd1hHus1 gene in cancer tissues and adjacent normal tissues, and to further analyze the difference between the expression levels and the TNM stage, pathological differentiation degree and location of the tumor. Sex, age and other clinicopathological data were related. The results showed that the expression of Rad9 m RNA in colorectal cancer tissues was significantly lower than that in adjacent normal tissues by using Spss17.0 software (P0.05). The expression of Rad9 mRNA in colorectal cancer tissues was 2.98 鹵5.14) lower than that in adjacent normal tissues (7.11 鹵16.82). The difference was statistically significant. There were 13 cases of poorly differentiated tumors with low expression of Rad9 mRNA and 9 cases with moderately differentiated tumors (64.29%). The low expression of Rad9 mRNA was found in 16 cases with lymph node metastasis (16 cases with lymph node metastasis) and 9 cases without lymph node metastasis (9 cases with lymph node metastasis).
