新疆哈薩克族ESCC組織LOXL2表達(dá)及臨床意義

【摘要】:目的哈薩克族食管癌屬于新疆地區(qū)特高發(fā)疾病,其發(fā)病率及死亡率均高于全國(guó)平均水平。近來(lái)越來(lái)越多的研究表明,賴氨酰氧化酶樣蛋白2(lysyl oxidase-like 2,LOXL2)與腫瘤的侵襲、轉(zhuǎn)移及不良預(yù)后有密切關(guān)系。本研究探討LOXL2在新疆哈薩克族食管鱗癌(esophageal squamous cell carcinoma,ESCC)患者中的表達(dá)與臨床病理特征相關(guān)性及與預(yù)后的關(guān)系。方法檢索Oncomine數(shù)據(jù)庫(kù)中LOXL2mRNA在ESCC患者中的表達(dá)情況,收集2009-03-01-2012-03-31新疆醫(yī)科大學(xué)附屬腫瘤醫(yī)院胸外科80例新疆哈薩克族ESCC患者手術(shù)切除標(biāo)本,免疫組織化學(xué)檢測(cè)LOXL2在80例新疆哈薩克族ESCC患者中蛋白表達(dá),并分析其表達(dá)與臨床病理特征及總生存時(shí)間(overall survival,OS)的關(guān)系。結(jié)果 Oncomine數(shù)據(jù)庫(kù)中共有2個(gè)研究涉及ECT2在ESCC組織和正常組織中mRNA表達(dá),2組微陣列數(shù)據(jù)集中,LOXL2mRNA在ESCC組織中的轉(zhuǎn)錄水平均顯著高于正常組織,P=3.95×10~(-16),P=6.87×10~(-7)。80例ESCC組織標(biāo)本及癌旁組織LOXL2蛋白陽(yáng)性表達(dá)率分別為55.0%和28.7%(χ2=4.665,P=0.031),且LOXL2蛋白表達(dá)與ESCC的腫瘤分化程度(χ~2=9.126、P=0.010)、淋巴結(jié)轉(zhuǎn)移(χ~2=4.393、P=0.036)及TNM分期(χ~2=6.519、P=0.011)呈正相關(guān),與患者性別、年齡、浸潤(rùn)深度及CEA水平無(wú)明顯相關(guān)性,P0.05。LOXL2陽(yáng)性表達(dá)陰性表達(dá)的患者5年OS分別為6.7%和35.8%,差異有統(tǒng)計(jì)學(xué)意義,χ~2=4.612,P=0.032。結(jié)論 LOXL2蛋白在新疆哈薩克族ESCC組織中表達(dá)明顯增高,其蛋白陽(yáng)性表達(dá)與腫瘤分化程度、淋巴結(jié)轉(zhuǎn)移及TNM分期密切相關(guān),LOXL2在新疆哈薩克族ESCC的發(fā)生發(fā)展中起重要作用,可以作為反映新疆哈薩克族食管癌生物學(xué)行為及預(yù)后的有效指標(biāo)。
[Abstract]:Objective the incidence and mortality of esophageal cancer in Kazak nationality are higher than the national average. More and more studies have shown that lysyl oxidase like protein 2lysyl oxidase-like 2LOXL2) is associated with tumor invasion. This study was designed to investigate the relationship between the expression of LOXL2 and clinicopathological features and prognosis in Xinjiang Kazak patients with esophageal squamous cell carcinoma. Methods the relationship between LOXL2mRNA and prognosis in Oncomine database was searched. Expression in patients, The specimens of 80 patients with ESCC of Kazak nationality in Xinjiang were collected from thoracic surgery department of Cancer Hospital affiliated to Xinjiang Medical University 2009-03-01-2012-03-31. The protein expression of LOXL2 was detected by immunohistochemistry in 80 cases of ESCC in Xinjiang Kazak nationality. The relationship between the expression of ECT2 and clinicopathological features and total survival time (OS) was analyzed. Results there were two studies on the expression of ECT2 in ESCC and normal tissues. Two groups of microarray data sets of mRNA expression in ESCC tissues were involved in this study. The positive expression rates of LOXL2 protein in ESCC tissues and adjacent tissues were 55.0% and 28.70.The expression of LOXL2 protein was positively correlated with the degree of differentiation of ESCC (蠂 ~ 29.126), lymph node metastasis (蠂 ~ 2 ~ 24.393P ~ (0.036)) and TNM stage (蠂 ~ (26.519) P 0.011), and the expression of LOXL2 protein was significantly higher than that of normal tissues (P ~ (3.95) 脳 10 ~ (-16)), and the positive expression rate of LOXL2 protein was significantly higher than that in normal tissues (蠂 ~ (2.95) 脳 10 ~ (-16)), and the positive rates of LOXL2 protein expression in ESCC tissues and adjacent tissues were 55.0% and 28.7 respectively (蠂 ~ (2) 4.665). The expression of LOXL2 protein was positively correlated with the degree of differentiation of ESCC (蠂 ~ 29.126). There was no significant correlation with sex, age, depth of infiltration and CEA level in the patients with negative expression of P0.05.LOXL2 in 5 years (P < 0.05). There was significant difference in the expression of LOXL2 protein in the ESCC tissues of Kazak nationality in Xinjiang, and the difference was statistically significant. Conclusion the expression of LOXL2 protein in the ESCC tissues of Kazakh nationality in Xinjiang is obviously higher than that in the control group (P < 0.05). The positive expression of Lox2 protein was closely related to tumor differentiation, lymph node metastasis and TNM staging, and played an important role in the occurrence and development of ESCC in Kazak nationality in Xinjiang. It can be used as an effective index to reflect the biological behavior and prognosis of Kazak esophageal cancer in Xinjiang.
