基于microcarrier 6構(gòu)建正常免疫小鼠人胃癌移植模型

【摘要】:目的:基于新型3D微載體(microcarrier 6)復(fù)合人胃癌MKN45細胞接種于具有正常免疫功能的小鼠,以期建立新型人胃癌正常小鼠轉(zhuǎn)移瘤模型。方法:將60只雄性C57BL/6小鼠按是否將MKN45細胞接種于支架材料隨機分為2D組、對照組和3D組;體外構(gòu)建三維腫瘤細胞培養(yǎng)模型,采用皮下接種法建立小鼠移植瘤模型,觀察小鼠成瘤時間、成瘤率、病理學(xué)表現(xiàn)等。結(jié)果:2D組和對照組均未成瘤,3D組成瘤率達80%,成瘤時間早,移植瘤HE染色和免疫組織化學(xué)均符合人胃癌特點。結(jié)論:本實驗成功基于microcarrier 6復(fù)合人胃癌MKN45細胞在正常免疫小鼠中建立人胃癌小鼠移植瘤模型,此模型可以更好地研究和進一步闡明胃癌在免疫功能正常機體中發(fā)生、發(fā)展機制,同時也為抗癌藥物的研發(fā)提供了更有價值的動物模型。
[Abstract]:Objective: to inoculate human gastric cancer MKN45 cells based on 3D microcarrier 6 in mice with normal immune function. Methods: 60 male C57BL / 6 mice were randomly divided into 2D group, control group and 3D group according to whether the MKN45 cells were inoculated into scaffold materials. The model of mouse transplanted tumor was established by subcutaneous inoculation. The tumorigenesis time, tumorigenesis rate and pathological manifestation of mice were observed. Both HE staining and immunohistochemistry were consistent with the characteristics of human gastric cancer. Conclusion: the transplanted tumor model of human gastric cancer mice was successfully established in normal immunized mice based on microcarrier 6 combined with human gastric cancer MKN45 cells. This model can better study and further elucidate the mechanism of gastric cancer in normal immune system and provide a more valuable animal model for the development of anticancer drugs.
