【摘要】：目的:通過研究突變型P53蛋白、Cyclin D1蛋白與宮頸癌組織學(xué)分級(jí)、病理類型、淋巴結(jié)轉(zhuǎn)移等病理特征之間相關(guān)性以及二者自身之間的關(guān)聯(lián),探討二者在宮頸癌發(fā)生發(fā)展過程中所具有的意義。方法:應(yīng)用免疫組化PV-6000二步法對(duì)2012-2015年青島大學(xué)附屬醫(yī)院18例正常宮頸組織、10例低級(jí)別宮頸上皮內(nèi)瘤變組織(CINI級(jí))、29例高級(jí)別宮頸上皮內(nèi)瘤變組織(CINII-III級(jí))、42例宮頸癌組織進(jìn)行免疫組織化學(xué)染色,檢測(cè)各組突變型P53及Cyclin D1蛋白表達(dá)情況。結(jié)果:(1)突變型P53蛋白在正常宮頸組、CINI組、CINII-III組、宮頸癌組中陽性表達(dá)率分別為0.00%、10.00%、44.82%、66.71%,呈明顯遞增趨勢(shì),差異有統(tǒng)計(jì)學(xué)意義(x2=27.763,P0.05)。Cyclin D1蛋白在正常宮頸組、CINI組、CINII-III組、宮頸癌組的陽性表達(dá)率分別為16.67%、20.00%、58.62%、83.33%,且差異有統(tǒng)計(jì)學(xué)意義(x2=34.270,P0.05)。(2)突變型P53和Cyclin D1蛋白在宮頸癌組織中的表達(dá)狀況均與組織分化程度、淋巴結(jié)有無轉(zhuǎn)移相關(guān)(P0.05),與病理類型、臨床分期無相關(guān)性(P0.05)。(3)在宮頸癌組織中突變型P53蛋白、Cyclin D1蛋白的陽性表達(dá)呈正相關(guān)(r=0.305,P0.05)。結(jié)論:突變型P53蛋白、Cyclin D1蛋白參與宮頸病變進(jìn)展,并與癌細(xì)胞分化程度及惡性程度有一定的關(guān)系,二者呈協(xié)同作用,共同促進(jìn)宮頸病變的進(jìn)展。
[Abstract]:Objective: to study the correlation between the mutant p53 protein Cyclin D1 protein and the histological grade, pathological type, lymph node metastasis and other pathological features of cervical cancer. To explore the significance of the two methods in the occurrence and development of cervical cancer. Methods: immunohistochemical PV-6000 two-step method was used to study 18 cases of normal cervix tissues in Qingdao University affiliated Hospital from 2012-2015 to 10 cases of low grade cervical intraepithelial neoplasia (CINI). 29 cases of high grade cervical intraepithelial neoplasia and 42 cases of cervical carcinoma of grade CINII-III were stained with immunohistochemical staining. Results the positive expression rates of p53 and Cyclin D1 protein in normal cervix cervix group and cervical carcinoma group were 0.0010.00 and 44.82%, respectively. There was significant difference in the expression of p53 and Cyclin D1 protein in normal cervix carcinoma group (CINI group) and cervical carcinoma group (P < 0.01). The positive expression rates of p53 and Cyclin D1 protein in cervical carcinoma group were 16.6767 ~ 20.00% and 88.62.33%, respectively, and the difference was statistically significant (P < 0.01) the expression of p53 and Cyclin D1 protein in cervical carcinoma tissues were all related to the degree of tissue differentiation. There was a positive correlation between the positive expression of mutant p53 protein and Cyclin D1 protein in cervical cancer tissues. Conclusion: the mutant p53 protein Cyclin D1 protein is involved in the progression of cervical lesions. And there is a certain relationship with the degree of differentiation and malignancy of cancer cells, both of which have synergistic effect to promote the progress of cervical lesions.
