【摘要】：嵌合抗原受體T細(xì)胞療法(CAR-T療法),是近年來發(fā)展迅速的一種新型細(xì)胞免疫療法。它通過基因重組和轉(zhuǎn)染技術(shù),獲得可以特異性地識(shí)別腫瘤相關(guān)抗原的效應(yīng)T細(xì)胞。與常規(guī)T細(xì)胞相比,CAR-T細(xì)胞具有更強(qiáng)的腫瘤靶向性、殺傷活性和持久性。隨著轉(zhuǎn)化醫(yī)學(xué)研究的不斷深入,CAR-T技術(shù)經(jīng)歷了4代優(yōu)化與革新,在眾多腫瘤疾病尤其是血液系統(tǒng)惡性腫瘤中展現(xiàn)出了有前景的臨床療效,但該技術(shù)的臨床應(yīng)用也帶來了脫靶效應(yīng)、細(xì)胞因子風(fēng)暴等風(fēng)險(xiǎn)。該文就CAR-T在血液腫瘤免疫治療中的進(jìn)展、不良反應(yīng)、應(yīng)對(duì)策略和發(fā)展前景作一綜述。
[Abstract]:Chimeric antigen receptor T cell therapy (CAR-T) is a new type of cellular immunotherapy developed rapidly in recent years. The effector T cells which can specifically recognize tumor-associated antigens were obtained. Compared with conventional T cells, CAR-T cells have stronger tumor targeting. With the development of transforming medicine, CAR-T technology has undergone four generations of optimization and innovation, and has shown a promising clinical effect in many tumor diseases, especially malignant tumors of the blood system. However, the clinical application of this technique also brings risks such as miss effect and cytokine storm. This article reviews the progress, adverse reactions, strategies and development prospects of CAR-T in immunotherapy of blood tumors.
